Single-cell sequencing resolves the landscape of immune cells and regulatory mechanisms in HIV-infected immune non-responders

Summary: Immune non-responder after highly active antiretroviral therapy (HAART) is a major problem in AIDS patients, leading to opportunistic infections and high mortality. The underlying mechanism of this immune reconstitution failure is poorly understood. In this study, single-cell sequencing and chromatin accessibility analysis were performed on peripheral blood mononuclear cells (PBMCs) from immune non-responders (INR) and responders (IR) with HIV-1 infection. The results showed that INRs had low expression of mucosal-associated invariant T (MAIT) cells, which were associated with impaired mitochondrial function and apoptosis signaling. Restoring mitochondrial function could potentially improve the immune dysfunction in MAIT cells against bacterial co-infections in INR subjects.
