Molecular Mechanisms Underlying IL-33-Mediated Inflammation in Inflammatory Bowel Disease

Ioanna Aggeletopoulou et al.

Summary: Interleukin-33 (IL-33) is a pleiotropic cytokine with both extracellular and nuclear functions, and its receptor ST2 plays a critical role in regulating inflammatory disorders. The IL-33/ST2 axis is involved in maintaining intestinal homeostasis and mucosal defenses. This review focuses on the pro- and anti-inflammatory effects of the IL-33/ST2 axis in ulcerative colitis and Crohn's disease, as well as the molecular mechanisms underlying its association with IBD pathogenesis. Understanding the IL-33/ST2-mediated mechanisms in IBD has potential for clinical applications as therapeutic targets.

Targeting interleukin-33 and thymic stromal lymphopoietin pathways for novel pulmonary therapeutics in asthma and COPD

Ariel A. Calderon et al.

Summary: Interleukin-33 (IL-33) and thymic stromal lymphopoietin (TSLP) are alarmins released upon airway epithelial injury that play critical roles in activating immune cell populations. They were believed to drive type 2 inflammation, but emerging evidence suggests their involvement in non-type 2 inflammation. Clinical trials have shown the potential of targeting IL-33 and TSLP in reducing exacerbations in asthma and COPD. Ongoing research focuses on these targets to further improve lung function and reduce exacerbations.

The alarmin interleukin-33 promotes the expansion and preserves the stemness of Tcf-1+CD8+T cells in chronic viral infection

Anna-Friederike Marx et al.

Summary: This study shows that T cell factor 1 (Tcf-1) expressing CD8+ T cells, known as stem-like CD8+ T cells (CD8+SL), play a crucial role in immune defense against chronic viral infection and cancer. It identifies interleukin-33 (IL-33) as a pivotal factor for the expansion and functioning of CD8+SL and virus control.

Interleukin-33 Mediates Cardiomyopathy After Acute Kidney Injury by Signaling to Cardiomyocytes

Nans Florens et al.

Summary: This study investigated the mechanism of cardiac remodeling induced by acute kidney injury (AKI) using mouse models. The results showed that IL-33 released from the kidney during AKI directly affected cardiac myocytes, leading to cardiac hypertrophy and cardiomyopathy. These findings suggest that antagonizing the IL-33/ST2 axis may have cardioprotective effects in patients with kidney disease.

Intestinal IL-33 promotes platelet activity or neutrophil recruitment during acute inflammation

Zuojia Chen et al.

Summary: Intestinal inflammation enhances platelet activation and blood coagulation, with intestinal epithelial damage leading to elevated levels of IL-33 and promotion of platelet activation through 5-HT release. Intestinal IL-33 and peripheral IL-33 have a protective role in acute inflammation.

Group 2 innate lymphoid cells (ILC2s) are important in typical type 2 immune-mediated diseases and an essential therapeutic target

Jie Jin et al.

Summary: The prevalence of allergic diseases has increased due to environmental changes. Type 2 immunity, particularly the ILC2-DC-Th2 axis, plays a crucial role in allergic diseases. ILC2s are activated by cytokines and lipid inflammatory mediators and produce a significant amount of type 2 cytokines. Therapeutic development targeting ILC2-derived cytokines is important in treating type 2-mediated diseases.
