RGS16 regulated by let-7c-5p promotes glioma progression by activating PI3K-AKT pathway

Summary: It has been confirmed that RGS16 is upregulated in glioma tissues and its high expression is associated with poor survival. Deleting RGS16 significantly suppresses glioma cell proliferation and migration. Moreover, RGS16 has been validated as a direct target gene of miR let-7c-5p. Therefore, the inhibition of RGS16 may promote glioma progression and could potentially serve as a prognostic biomarker and therapeutic target for glioma.

Summary: The study discovered that Hypoxia-Inducible Factor-1 alpha is up-regulated in clinical preeclampsia samples, while Regulator of G Protein Signaling 2 is down-regulated. Mechanistically, Hypoxia-Inducible Factor-1 alpha is induced in response to hypoxia, forming a complex with E1A binding protein P300 that inhibits the transcription of Regulator of G Protein Signaling 2, potentially contributing to the development of preeclampsia.

Low Expression of RGS2 Promotes Poor Prognosis in High-Grade Serous Ovarian Cancer

Summary: Recent advances in molecular medicine have highlighted the potential therapeutic targets of G-protein coupled receptors (GPCRs) in ovarian cancer, particularly the regulator of G protein signaling 2 (RGS2). This study retrospectively analyzed the expression and prognostic value of RGS2 in high-grade serous ovarian cancer (HGSOC). The results showed that low protein expression of RGS2 was associated with inferior overall survival and progression-free survival. Further exploration is needed to understand the functional role of RGS2 in HGSOC and its implications for new drug discovery.

Summary: Experimental evidence in mice models has shown that high levels of the regulator of G-protein signaling 2 (RSG2) protein are associated with insulin resistance, and in patients with type 2 diabetes mellitus, platelet RGS2 expression levels were higher than in healthy donors. Overexpression of RGS2 in human endothelial cells leads to decreased insulin signaling and is linked to poorly controlled diabetes.

Mechanistically, miR-3935 inhibits TRAF6, which reduces its inhibition on RGS2, leading to decreased methylation of the CDH1 gene promoter through ALKBH1 induction, subsequently increasing E-Cadherin and causing insufficient trophoblast EMT.
