High spatial resolution mass spectrometry imaging for spatial metabolomics: Advances, challenges, and future perspectives

Mass spectrometry imaging (MSI) technologies enable direct visualization of chemical distributions in biological samples without labeling and are gaining popularity for biological applications. Spatial metabolomics and MSI generate extensive visualization information, extending metabolomics from point to plane and driving the development of high spatial resolution MSI and its application in spatial metabolomics.

Nano-DESI Mass Spectrometry Imaging of Proteoforms in Biological Tissues with High Spatial Resolution

Summary: Mass spectrometry imaging (MSI) is a powerful technique for mapping the spatial distribution of proteins in biological tissues. In this study, we demonstrate proteoform mapping in biological tissues using nano-DESI MSI, achieving a spatial resolution down to 7 μm. We have improved the sensitivity of nano-DESI MSI experiments by optimizing the design of the capillary-based probe and the thickness of the tissue section. These developments represent a new step in MSI-based spatial proteomics and complement targeted imaging modalities used for studying biological systems.

High-throughput mass spectrometry imaging of biological systems: Current approaches and future directions

Summary: In the past two decades, the power of mass spectrometry imaging (MSI) for label-free spatial mapping of molecules in biological systems has been substantially enhanced through the development of high spatial resolution imaging approaches. However, the experimental throughput has become a limiting factor for imaging large samples and 3D imaging of tissues. To address this issue, several experimental and computational approaches have been recently developed to improve the throughput of MSI. These approaches focus on speeding up sampling, reducing mass spectrometer acquisition time, and minimizing the number of sampling locations. The rate-determining steps for different MSI methods and future directions in the development of high-throughput MSI techniques are discussed.

MALDI TIMS IMS of Disialoganglioside Isomers?GD1a and GD1b in Murine Brain Tissue

Summary: Gangliosides are diverse acidic glycosphingolipids with distinct biological roles. Matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI IMS) can be used for spatial analysis of gangliosides directly from tissue sections. The integration of trapped ion mobility spectrometry with MALDI IMS enables the analysis of isomeric lipid structures in situ. Gas-phase separation and identification of disialoganglioside isomers GD1a and GD1b, with different sialic acid residue positions, were demonstrated in multiple samples, including tissue sections.

Tandem Mass Spectrometry Imaging Enables High Definition for Mapping Lipids in Tissues

Summary: Mass spectrometry imaging (MSI) of lipids in biological tissues can provide valuable information for biological research and disease diagnosis by correlating molecular distribution with pathological results. A new multiplexed MS2 imaging method (MSI-I-2) has been developed, allowing for MS2 images of 10 or more lipid species at each sampling spot, providing spatial structural lipidomic information. Coupled with on-tissue photochemical derivatization, imaging of 20 phospholipid C=C location isomers is also possible, resulting in enhanced molecular images with high structural definition for mouse brain and human liver cancer tissue sections. Spatially mapped t-distributed stochastic neighbor embedding has been utilized to visualize the tumor margin with the assistance of structural lipidomic information.

Enhancement of lipid signals with ammonium fluoride in negative mode Nano-DESI mass spectrometry imaging
