Unravelling the mechanisms of drugs partitioning phenomena in micellar systems via NMR spectroscopy

Summary: In this study, the interaction between small guest molecules and surfactants forming micelles was investigated using NMR techniques and molecular dynamics simulations. The molecular organization of drugs within the hydrophilic and hydrophobic domains of micellar aggregates was revealed, and it was found that flexible surfactants do not follow the classical core-shell model of micelles. These findings have important implications for the analysis of host-guest systems in a broad range of colloidal systems, from soft materials to biological systems.

Efficient pKa Determination in a Nonaqueous Solvent Using Chemical Shift Imaging

Summary: pK(a) is an important property of a molecule that has significant applications in drug design, catalysis, reactivity, and environmental toxicity. Existing NMR methods for measuring pK(a) in water are well established, but measuring pK(a) in organic solvents is laborious and problematic. This paper presents an efficient one-shot method to determine the pK(a) of a molecule in an organic solvent by establishing a pH gradient in the NMR tube.

Does It Bind? A Method to Determine the Affinity of Calcium and Magnesium Ions for Polymers Using 1H NMR Spectroscopy

Summary: The binding of calcium and magnesium ions (M2+) by polymers and other macromolecules in aqueous solution is a common phenomenon in chemistry and biology. A new method based on H-1 NMR chemical shift imaging (CSI) is presented, which allows for the assessment of the binding affinity of polymers using standard high-field NMR equipment in a single experiment.

Mechanistic Influence of Polymer Species, Molecular Weight, and Functionalization on Mucin-Polymer Binding Interactions

Summary: Optimizing delivery vehicle properties is crucial for improving the delivery of pharmaceuticals. Mucoadhesion, a designed property that enhances the interaction between polymers and mucins, is an effective approach to increase drug residence times and uptake efficiency. Using nuclear magnetic resonance, this study investigated the influence of polymer design parameters on mucoadhesion and uncovered key findings on the polymer-mucin interaction at the atomic level. The results provide insights into the importance of polymer design and mucoadhesive functional groups for drug delivery.
