Electrochemiluminescence Distance and Reactivity of Coreactants Determine the Sensitivity of Bead-Based Immunoassays

Yafeng Wang et al.

Summary: In this study, the electrochemiluminescence (ECL) generation by Ru(bpy)(3)(2+) and five tertiary amine coreactants was investigated. The ECL distance and reactivity of the coreactants were measured, and the integrated ECL intensity was used to quantitatively evaluate the reactivity. Statistical analysis of ECL images of labeled microbeads showed that the ECL distance and reactivity of the coreactant determine the emission intensity and sensitivity of immunoassay. BIS-TRIS was found to balance the trade-off between ECL distance and reactivity, resulting in a 236% increase in sensitivity compared to TPrA in the bead-based immunoassay of carcinoembryonic antigen.

An electrochemiluminescence imaging sensor for the analysis of lactate in foods via a single gold microsphere

Gen Liu et al.

Summary: To address concerns about food safety, a fast and high-throughput lactate determination method is urgently needed. In this study, a novel electrochemiluminescence (ECL) imaging sensor based on a single gold microsphere with lactate oxidase (LOD) was developed. The sensor achieved sensitive detection of lactate by visually measuring the ECL intensity at a single gold microsphere. It exhibited a good linear relationship in the range of 0.1-50 mM of lactate, with a detection limit of 0.02 mM (S/N = 3).

Single Cell Imaging of Electrochemiluminescence-Driven Photodynamic Therapy

Ming-Ming Chen et al.

Summary: We report a photodynamic therapy driven by electrochemiluminescence (ECL) with potential clinical applications. The ECL-generated luminescence acts as both optical readout and light source for the excitation of photosensitizer to produce reactive oxygen species (ROS). Real-time monitoring of cellular processes during ECL-PDT was achieved using ECL microscopy with high spatiotemporal resolution.

A redox-mediator pathway for enhanced multi-colour electrochemiluminescence in aqueous solution

Emily Kerr et al.

Summary: A novel 'redox mediator' pathway utilizing fac-[Ir(sppy)(3)](3-) has been discovered to significantly enhance the ECL reaction of [Ru(bpy)(3)](2+) and tri-n-propylamine. The green emission of [Ir(sppy)(3)](3-)* allows for internal standardization of the reaction, enabling superior sensitivity and precision for low-cost, portable ECL-based analytical devices when using a digital camera as the photodetector.

Spatially Selective Imaging of Cell-Matrix and Cell-Cell Junctions by Electrochemiluminescence

Hao Ding et al.

Summary: A novel method for spatially selective imaging of cell junctions by regulating the thickness of the electrochemiluminescence (ECL) layer through control of luminophore and co-reactant concentrations has been reported. Low luminophore concentration reveals cell-matrix adhesions, while high concentration allows sequential imaging of cell-matrix and cell-cell junctions. This strategy not only provides new insights into ECL mechanisms but also expands the potential applications of ECL microscopy in bioimaging.
