Ion Conduction Mechanisms in Potassium Channels Revealed by Permeation Cycles

Summary: We use molecular dynamics simulations and Markov state models to investigate the dynamical details of ion permeation in potassium channels. Our results reveal that the direct knock-on permeation is the dominant mechanism across various conditions, and it is also observed in other potassium channels with conserved selectivity filters. Furthermore, we explore the dependence of permeation cycles on charge strength. These findings provide valuable insights into the conduction mechanisms in potassium channels.

Voltage-Dependent FTIR and 2D Infrared Spectroscopies within the Electric Double Layer Using a Plasmonic and Conductive Electrode

Summary: Strong electric fields between electric double layer and charged surfaces impact molecular structures and chemistry at interfaces. A transparent electrode with infrared plasmonic enhancement has been developed to measure FTIR and two-dimensional infrared spectra at submonolayer coverages on a surface with applied voltage. The electrode design, optimized through material characterization and spectroscopy, allows for voltage-dependent infrared experiments.

Voltage-sensor movements in the Eag Kv channel under an applied electric field

Structure Changes of a Membrane Polypeptide under an Applied Voltage Observed with Surface-Enhanced 2D IR Spectroscopy

Summary: The study demonstrates that voltage-dependent structures of membrane-bound polypeptide alamethicin can be measured using a spectroelectrochemical cell with a rough gold film, revealing changes in structure under different voltage and pH conditions. Ion Conduction Mechanism as a Fingerprint of Potassium Channels

Summary: Through molecular dynamics simulations and Markov state models, a comparative study of ion conduction in three distinct channel models revealed significant differences in terms of permeation mechanisms and binding site occupancy. Each system was found to display a characteristic permeation mechanism, challenging the current paradigm that there is a unique way by which potassium ions move through K+-channels. The diversity in conduction features identified in this study may contribute to the high functional diversity of K+-channels.
