Atlas of type 2 dopamine receptors in the human brain: Age and sex dependent variability in a large PET cohort

Summary: This study investigated the influence of age, sex, BMI, cerebral hemisphere, and regional volume on striatal D2R availability in the human brain using a large historical dataset. The results showed that D2R availability decreased with age and was higher in females than males. BMI was weakly associated with D2R availability. The observed effects were independent of regional volumes and were validated using different spatial normalization methods. These findings contribute to our understanding of the vulnerability of neurological and psychiatric conditions involving altered D2R expression.

Reproducing the dopamine pathophysiology of schizophrenia and approaches to ameliorate it: a translational imaging study with ketamine

Summary: Schizophrenia patients show increased striatal dopamine synthesis capacity, possibly due to NMDAR hypofunction and PV neuronal dysfunction. A mouse model mimicking this dopamine pathology was developed, with sub-chronic ketamine treatment increasing dopamine synthesis capacity and locomotor activity. Activation of midbrain dopamine neurons and PV interneurons reversed these effects, indicating potential therapeutic targets.

Summary: This meta-analysis evaluated the efficacy of ketamine for major depressive episodes and found significant treatment effects at 2, 4, and 6-weeks post-treatment. The results suggest that ketamine can be an effective pharmacologic intervention with lasting effects up to 6 weeks post-administration, which is promising for treatment outcomes.
