Cytotoxic activity of nitrogen, sulfur, and oxygen chelated Pt(II) complexes; their DNA/BSA binding by in vitro and in silico approaches

Summary: A series of Pt(II) complexes with nitrogen, sulfur, and oxygen donors were synthesized and evaluated for their anticancer activity. The theoretical calculations supported the experimental findings, and various techniques were used to analyze the binding modes and binding aptitude of the complexes. The results showed that these complexes exhibited in vitro anticancer activity against breast cancer cells and induced cell death through ROS generation.

Time-Dependent Studies of Oxaliplatin and Other Nucleolar Stress-Inducing Pt(II) Derivatives

Summary: The properties of small molecule Pt(II) compounds that drive specific cellular responses are of interest due to their broad clinical use as chemotherapeutics as well as to provide a better mechanistic understanding of bioinorganic processes. Different Pt(II) derivatives exhibit variations in rates and degrees of nucleolar stress induction and inhibition of rRNA transcription, potentially due to the ring size and stereochemistry of the non-aquation-labile ligand. The onset and overall degree of nucleolar stress are faster and higher in compounds with a 6-membered ring and 1R,2R-stereoisomeric conformation.

The synthesized esters exhibited significant antibacterial and antifungal activities, and some of them showed inhibitory effects on cancer cells. Molecular docking and molecular dynamics simulations confirmed the binding affinities and interactions between the compounds and pathogen proteins. In addition, pharmacokinetics prediction and drug-likeness analysis indicated that the compounds possessed favorable drug properties.

Furthermore, Pt1 complex induced apoptosis in 4T1 and A549 cells by increasing pro-apoptotic caspase-3 expression and decreasing anti-apoptotic Bcl-2 and Ki-67 expression, suggesting its potential antitumor capacity through facilitating apoptosis and inhibiting tumor cell proliferation.
