Monitoring polysorbate hydrolysis in therapeutic proteins using an ultrasensitive extraction-free fatty acid quantitation method

Sisi Zhang et al.

Summary: A rapid lipase activity assay was introduced for accurate quantitation of free fatty acids without the need for a fatty acid extraction step. This assay can detect low levels of PS degradation within 1 day and the released free fatty acids correlate strongly with the degree of PS20 degradation.

Quantitative Evaluation of Insoluble Particulate Matters in Therapeutic Protein Injections Using Light Obscuration and Flow Imaging Methods

Hiroko Shibata et al.

Summary: This study compared the effectiveness of flow imaging (FI) and light obscuration (LO) methods in evaluating insoluble particles in therapeutic protein injections. The results showed that FI method yielded higher particle counts compared to LO method, and the difference varied depending on the products or features of particles.

An Intercompany Perspective on Compatibility and In-use Stability Studies to Enable Administration Of Biopharmaceutical Drug Products

Douglas E. Kamen et al.

Summary: This article discusses an inter-company collaboration exercise conducted by the Formulation Workstream of the BioPhorum Development Group (BPDG) to understand current industry approaches for in-use stability and compatibility studies of biologic drugs. The survey results provide insights into common industry practices and offer various approaches to guide the design of an in-use stability and compatibility program based on clinical and biomolecule needs.

Human Serum Albumin Mitigates Formation of Fatty Acid Particles in Polysorbate-Containing Solutions

Dorothy M. Kim et al.

Summary: Monoclonal antibody therapeutics often contain polysorbate for stabilizing effects, but this may lead to particle formation. The presence of human serum albumin can prevent and dissolve fatty acid particles, providing a plausible explanation for previous observations and reducing concerns about particle levels in the final drug product.

Dissolution of Polysorbate 20 Degradation Related Free Fatty Acid Particles in Intravenous Bag Solutions

Nidhi Doshi et al.

Summary: The degradation of Polysorbate 20 (PS20) in drug product formulations can lead to the generation of free fatty acids (FFAs) which can form visible and/or sub-visible particles. However, the FFAs particles dissolve immediately upon dilution in intravenous solutions, reducing the risk of these particles being administered to a patient intravenously.

In-Use Interfacial Stability of Monoclonal Antibody Formulations Diluted in Saline i.v. Bags

Aadithya Kannan et al.

Summary: The use of monoclonal antibodies (mAbs) for disease treatment is increasing rapidly, with a higher aggregation propensity observed in saline compared to low ionic strength formulation buffer. The presence of salt in buffer with surfactants leads to greater mAb adsorption at the interface and the formation of more particulate aggregates. Stress studies for mAb formulations intended for intravenous administration should be performed in conditions representative of their intended route of administration.

Extended Characterization and Impact of Visible Fatty Acid Particles - A Case Study With a mAb Product

Miguel Saggu et al.

Summary: This study presents a case analysis of visible particles in biopharmaceutical formulations, specifically focusing on particles resulting from the degradation of the surfactant polysorbate 20. Through techniques such as mass spectrometry, Raman spectroscopy, and flow imaging microscopy, the identity and composition of the particles were determined to support risk assessments. The impact of saline and human plasma exposure on the fate of these particles upon dilution and/or administration was also evaluated to better understand their behavior in clinical settings.

Robert G. Strickley et al.

Summary: This review examines 126 commercially available antibodies approved globally between 1986 and February 2021, including various formulations, routes of administration, and formulation components. The number of approved antibodies per year has increased significantly after 2014. Most antibody products are packaged in single-dose units, with different routes of administration and formulation components.

Polysorbate Degradation and Particle Formation in a High Concentration mAb: Formulation Strategies to Minimize Effect of Enzymatic Polysorbate Degradation

Ian Roy et al.

Summary: This manuscript summarizes the investigation of a batch of high concentration (>100 mg/mL) mAb DP where subvisible particles formed abruptly after prolonged storage at 5°C degrees. The concentration and fatty acid composition of polysorbates were found to be significant factors in particle development. Solubilizers and alternative surfactants were all shown to be effective means of preventing particle formation.

Stability of Biologics and the Quest for Polysorbate Alternatives

Sachin Dubey et al.

Summary: Most biopharmaceutical formulations currently rely on polysorbates, which are highly efficient but challenging to control in terms of quality and stability. However, alternatives such as poloxamers, albumin, and cyclodextrin are gaining popularity for their potential to protect biopharmaceuticals against physical and mechanical stresses.
