Comprehensive investigation on the metabolism of emodin both in vivo and in vitro

Summary: This study comprehensively investigated the absorption, metabolism, distribution, and excretion of emodin. The absolute bioavailability of emodin was found to be approximately 3.2%, with around 56% of emodin remaining unabsorbed and excreted mainly in feces. Metabolites of emodin, including hydroxylated and glucuronidated metabolites, were rapidly formed in the body. The distribution of emodin and its metabolites was mainly observed in the kidney, while excretion occurred predominantly through urine and feces for hydroxylated metabolites, and through urine and bile for glucuronidated metabolites. The study also identified key liver enzymes involved in the metabolism of emodin, namely CYP1A2, CYP2E1, UGT1A1, UGT1A9, and UGT2B7. This comprehensive study provides valuable insights into the pharmaceutical and toxicological effects of emodin.

Development and validation of LC-MS/MS methods for the determination of EVT201 and its five metabolites in human urine: Application to a mass balance study

Summary: In this study, two LC-MS/MS methods were developed and validated to quantify EVT201 and its metabolites in human urine for the treatment of insomnia. The methods showed good selectivity, accuracy, precision, and were successfully applied to a mass balance study. The results indicated that EVT201 had high oral bioavailability and urinary elimination was its main excretion pathway in humans.

Targeting tryptophan metabolism reveals Clematichinenoside AR alleviates triptolide-induced hepatotoxicity

Summary: Liver toxicity induced by Triptolide has limited its clinical application on rheumatoid arthritis. Saponins have been proved as an efficacious remedy to mitigate hepatotoxicity. Tryptophan metabolites activate transcriptional regulators to mediate host detoxification responses. Research aimed to investigate whether Clematichinenoside AR could attenuate TP-induced liver damage by regulating Trp metabolism. Therapeutic modulation with natural compounds might be a useful clinical strategy to ameliorate toxicity induced by TP. Deciphering Trp metabolism will facilitate a better understanding of the pathogenesis of diseases and drug responding.
