Morphological distribution mapping: Utilisation of modelling to integrate particle size and shape distributions

John F. Gamble et al.

Summary: The aim of this work was to develop approaches to utilize whole particle distributions for both particle size and particle shape parameters to map the full range of particle properties in a curated dataset. A 1-dimensional principal component analysis (PCA) approach was applied to create a 'morphological distribution landscape'. The curated dataset encompassed over 200 different materials, which included over 150 different APIs, and approximately 3500 unique samples.

Impact of Crystal Nuclei on Dissolution of Amorphous Drugs

Xin Yao et al.

Summary: Amorphous drugs are used to improve the bioavailability of poorly water-soluble drugs. However, the presence of crystal nuclei in amorphous films can significantly negate or eliminate the dissolution advantage of amorphous drugs. Understanding the impact of crystal nucleation on the dissolution of amorphous drugs is important in order to fully realize the benefits of amorphous solids.

Elucidating the Dehydration Mechanism of Nitrofurantoin Monohydrate II Using Low-Frequency Raman Spectroscopy

Peter J. G. Remoto et al.

Summary: The dehydration of nitrofurantoin monohydrate II was studied using Raman spectroscopy at different isothermal conditions. Clear and subtle differences in the Raman spectra were observed for solid-state transformations. Multivariate curve resolution and theoretical modeling were used to analyze the solid-state forms and their relative abundances. Mid-frequency Raman spectroscopy detected the onset of the dehydration process earlier than low-frequency Raman spectroscopy.
