Flavescence doree phytoplasma enters insect cells by a clathrin-mediated endocytosis allowing infection of its insect vector

Nathalie Arricau-Bouvery et al.

Summary: To infect its insect vector, the flavescence doree phytoplasma invades different cell types using clathrin-mediated endocytosis. In this study, we found that the phytoplasmas could enter insect cells via clathrin-mediated endocytosis by interacting with the adaptor protein complexes AP-1 and AP-2. Inhibition of clathrin activity or knockdown of clathrin heavy chain gene expression decreased phytoplasma entry into insect cells, confirming the importance of clathrin in the colonization of the insect vector.

Silencing of ATP Synthase β Impairs Egg Development in the Leafhopper Scaphoideus titanus, Vector of the Phytoplasma Associated with Grapevine Flavescence Doree

Matteo Ripamonti et al.

Summary: This study discovered the presence of RNA interference (RNAi) machinery in the insect species Scaphoideus titanus, which is the natural vector of a plant disease. Injection of dsRNA targeting a specific gene in S. titanus led to gene silencing and observed changes in egg development and protein expression.

Leafhopper feeding behaviour on three grapevine cultivars with different susceptibilities to Flavescence dor acute accent ee

Matteo Ripamonti et al.

Summary: The feeding behavior of Scaphoideus titanus on different grape varieties was studied, revealing differences in the duration of the phloem feeding phase and the frequency of interruption-salivation events. The preference of the leafhopper for the Barbera cultivar correlated with an enhanced transmission efficiency of FD, explaining the higher susceptibility of this variety to the disease.

Metatranscriptomic Assessment of the Microbial Community Associated With the Flavescence doree Phytoplasma Insect Vector Scaphoideus titanus

Simona Abba et al.

Summary: Phytoplasmas are pathogenic bacteria transmitted by insects, causing significant economic losses in crops worldwide. The microbial community associated with insect vectors plays a role in their biology and vector competence. Studying the diversity of the microbiome of phytoplasma insect vectors is important for developing microbe-based pest control strategies and reducing the use of insecticides. This study used RNA-Seq data to analyze the microbial community associated with the leafhopper Scaphoideus titanus in its native and invasive areas. The findings identified stable microbial associations and highlighted potential targets for biocontrol strategies against Flavescence doree spread in Europe.

Foliar application of clay-delivered RNA interference for whitefly control

Ritesh G. Jain et al.

Summary: Foliar spray of double-stranded RNA carried on clay particles or BioClay can effectively disrupt multiple life stages of whitefly in crop hosts. The use of adjuvants enhances the uptake and movement of foliar-applied dsRNA. By delivering the dsRNA as a BioClay spray, it improves protection against immature insect stages and shows potential for complete life cycle control of whitefly and possibly other pests.

RNAi by Soaking Aedes aegypti Pupae in dsRNA

Fiza Arshad et al.

Summary: RNA-interference (RNAi) is a standard technique for functional genomics in adult mosquitoes, but its application in immature stages has been challenging. This study successfully developed a simple method for RNAi in mosquito pupae by soaking them in water containing dsRNA, without the need for carrier molecules or microinjections. The technique can be used to study gene expression in pupae and potentially identify new targets for mosquito control at early life stages.
