Repolarization of Unbalanced Macrophages: Unmet Medical Need in Chronic Inflammation and Cancer

Yannick Degboe et al.

Summary: Monocytes and macrophages play a crucial role in the immune system, detecting and eradicating danger signals while maintaining tissue homeostasis. Imbalance in their response can lead to pathological disorders, and repolarization of these unbalanced macrophages shows promise as a therapeutic strategy.

Innate Phagocyte Polarization in the Oral Cavity

Sarah Metcalfe et al.

Summary: This review focuses on the roles of macrophages and neutrophils in oral homeostasis and disease development, particularly in periodontal disease and cancer. The polarization changes of macrophages play a crucial role in maintaining oral health and contributing to disease progression. Recent studies also reveal phenotypic heterogeneity in neutrophils within the oral cavity, and their interactions with oral microbiota can worsen disease states.

Functional Hallmarks of Healthy Macrophage Responses: Their Regulatory Basis and Disease Relevance

Katherine M. Sheu et al.

Summary: This review discusses the importance and functions of macrophages in the immune system. The effector and sentinel functions of healthy macrophages rely on response specificity, context dependence, and stimulus memory. Impairment of these hallmark properties may lead to immune dysregulation.

Immunoglobulin gene expression profiles and microbiome characteristics in periodontitis in nonhuman primates

Jeffrey L. Ebersole et al.

Summary: This study using a nonhuman primate model suggests that the formation of periodontal lesions is closely related to adaptive immune responses and that age affects the expression of Ig genes, which may be related to the increased prevalence and severity of periodontitis in older individuals.

Oral microbiome interactions with gingival gene expression patterns for apoptosis, autophagy and hypoxia pathways in progressing periodontitis

Jeffrey L. Ebersole et al.

Summary: This study investigated the interaction between the oral microbiome and transcriptomic footprints of apoptosis, autophagy, and hypoxia pathways during periodontitis. The results showed significant correlations between altered gene expression profiles in apoptosis, autophagy, and hypoxia in both health and disease samples. Bacterial complexes were found to correlate significantly with host gene profiles in different stages of health, disease, and resolution. The data suggest dysbiosis at the functional genomic level with different interactions between bacteria and host cells in healthy and diseased conditions.

Cell Type-Specific Decomposition of Gingival Tissue Transcriptomes

F. Momen-Heravi et al.

Summary: Genome-wide transcriptomic analyses in whole tissues may not accurately identify cell type-specific information. This study utilized computational methods to decompose gene expression in gingival tissues into cell type-specific signatures, identifying differentially expressed genes and dysregulated pathways in periodontitis. Validation of these findings highlighted the potential roles of these genes in the pathogenesis of periodontitis.

Age and Sex: Impact on adipose tissue metabolism and inflammation

Mita Varghese et al.

Summary: Age-associated chronic inflammation plays a significant role in diseases as individuals age, with adipose tissue function being central to metabolic disease and longevity. Hormonal fluctuations in aging may regulate age-related visceral adiposity and metabolic dysfunction. It is important to consider sex differences in macrophage polarization, changes in T and B cell numbers, and dendritic cell types in relation to age-related inflammatory phenomena.

Summary: Macrophages play crucial roles in periodontitis, with different types impacting the immune system and the destruction and repair of periodontal tissues. Polarized M1 and M2 macrophages are involved in inflammatory and anti-inflammatory stages respectively, regulating the progression of periodontitis.

Transcriptomic phases of periodontitis lesions using the nonhuman primate model

Jeffrey L. Ebersole et al.

Summary: By studying the transcriptomic changes in a nonhuman primate model of ligature-induced periodontitis, researchers identified distinct patterns of gene expression in gingival tissue during different phases of the disease process. These findings contribute to a better understanding of the biological dynamics of periodontitis and may potentially enhance patient management through detection of specific gene products in gingival crevicular fluid samples.

Macrophages: The Good, the Bad, and the Gluttony

Ewan R. A. Ross et al.

Summary: Macrophages play critical roles in sterile inflammation and autoimmune diseases, particularly in the pathophysiology of diseases like rheumatoid arthritis. Imbalance in macrophage homeostasis contributes to chronic inflammation, and therapeutic strategies targeting macrophage plasticity are being explored. Apoptotic cell-derived extracellular vesicles may play a role in regulating macrophage phenotype and are a potential target for new therapeutic approaches.
