NAP1L2 drives mesenchymal stem cell senescence and suppresses osteogenic differentiation

Meilin Hu et al.

Summary: The elevation of NAP1L2 expression is correlated with the senescence and impaired osteogenesis of BMSCs, and NAP1L2 regulates BMSC cell senescence and osteogenic differentiation through NF-kappa B pathway, H3K14ac modification, and chromatin accessibility. Additionally, there is a potential interaction between NAP1L2 and the anti-aging reagent NMN, which alleviates senescent phenotypes of BMSCs. These findings provide a new theoretical basis for aging-related diseases.

MiR-152-5p suppresses osteogenic differentiation of mandible mesenchymal stem cells by regulating ATG14-mediated autophagy

Shaoming Li et al.

Summary: This study identified miR-152-5p as an important regulator in osteogenic differentiation by directly targeting autophagy-related protein ATG14. Inhibition of miR-152-5p promoted osteogenic differentiation by enhancing autophagy and reducing oxidative stress. These findings suggest that therapeutic inhibition of miR-152-5p may be an effective strategy for the treatment of osteoporosis.

S100A11 Promotes Liver Steatosis via FOXO1-Mediated Autophagy and Lipogenesis

Linqiang Zhang et al.

Summary: The study revealed that S100A11 was significantly up-regulated in NAFLD, and its overexpression could induce hepatic lipid accumulation by activating autophagy and lipogenesis processes. S100A11 may interact with HDAC6 to block its binding to FOXO1, resulting in the activation of autophagy and lipogenesis, thereby promoting lipid accumulation and liver steatosis. These findings suggest a novel S100A11-HDAC6-FOXO1 axis in regulating autophagy and liver steatosis, providing potential therapeutic targets for NAFLD.

Oral microbiome interactions with gingival gene expression patterns for apoptosis, autophagy and hypoxia pathways in progressing periodontitis

Jeffrey L. Ebersole et al.

Summary: This study investigated the interaction between the oral microbiome and transcriptomic footprints of apoptosis, autophagy, and hypoxia pathways during periodontitis. The results showed significant correlations between altered gene expression profiles in apoptosis, autophagy, and hypoxia in both health and disease samples. Bacterial complexes were found to correlate significantly with host gene profiles in different stages of health, disease, and resolution. The data suggest dysbiosis at the functional genomic level with different interactions between bacteria and host cells in healthy and diseased conditions.

Extracellular IL-37 promotes osteogenic and odontogenic differentiation of human dental pulp stem cells via autophagy

Na Li et al.

Summary: IL-37 enhances the osteogenic and odontogenic differentiation ability of dental pulp stem cells (DPSCs) without affecting their proliferation, and also activates cell autophagy. Activation of autophagy promotes osteogenesis and odontogenesis of DPSCs, while inhibition of autophagy suppresses their differentiation.

Chloroquine-Induced Accumulation of Autophagosomes and Lipids in the Endothelium

Ewelina Bik et al.

Summary: The antimalarial drug Chloroquine (CQ) was found to not be toxic to endothelial cells at concentrations of 1-30 μM and did not induce inflammation. However, it did lead to increased staining intensity of neutral lipids, lysosomotropism, and autophagy inhibition. This indicates alterations in lipid composition of endothelial cells upon exposure to CQ.

HDAC6 inactivates Runx2 promoter to block osteogenesis of bone marrow stromal cells in age-related bone loss of mice

Chao Ma et al.

Summary: Senile osteoporosis is a common and severe disease affecting the elderly, characterized by increased risk of fractures due to bone fragility. Research focused on investigating the mechanisms underlying defective bone formation in age-related bone loss, particularly the role of histone acetylation. Inhibition of HDAC6 was found to improve osteogenic potential in aged mice, providing insights for potential therapies to treat senile osteoporosis.
