α-Benzylation of Carbonyl Compounds Enabled by Synergistic Copper/Amine Catalyzed Decarboxylation of Arylacetic Acids

Summary: In this study, synergistic copper/amine-catalyzed reactions were used to directly a-benzylate carbonyl compounds with arylacetic acids. These reactions demonstrate the first examples of decarboxylative oxidative synergistic transition-metal/enamine catalysis for a-functionalizing carbonyl compounds. The process is characterized by availability of starting materials, good tolerance towards functional groups, mild reaction conditions, and high yields. Dibenzylated products were obtained in moderate yields under slightly modified reaction conditions, and some products underwent acid-catalyzed intermolecular cyclization reactions with indoles to form 1,2,3,4-tetrahydrocyclopenta[b]indole derivatives. Furthermore, biological studies revealed that some of the products possess anticancer properties and may be beneficial for drug discovery studies.

Copper-Mediated C4-Benzylations of 5-Aminopyrazoles with 3-Indoleacetic Acids

Summary: In this study, a copper-mediated C4-benzylation reaction of 5-aminopyrazoles with 3-indoleacetic acids was presented. Various benzylated 5-aminopyrazoles were synthesized in good-to-excellent yields under basic and ligand-free conditions in the presence of copper acetate. The benzylation method was also applied to other substrates and showed potential antiproliferative activities against cancer cell lines. Moreover, the C4-benzylated products underwent cyclization with aldehydes to form 1H-pyrazolo-[4 ',3 ':6,7]-azepino-[3,4-b]-indoles via one-pot two-step processes. The cyclized products exhibited fluorescence emissions with large Stokes shifts.

Diversifying Amino Acids and Peptides via Deaminative Reductive Cross-Couplings Leveraging High-Throughput Experimentation

Summary: A deaminative reductive coupling method was developed for the efficient synthesis of noncanonical amino acids and diversification of peptides. The method allows the transformation of natural and commercially available amino acids to aryl alanines and derivatives with varying chain lengths. It has attractive features such as scalability, tolerance of pharma-relevant aryls and biorthogonal functional groups, and applicability to short and macrocyclic peptide substrates.

N-Heterocyclic Carbene-Catalyzed Atroposelective Synthesis of Pyrrolo[3,4-b]pyridines with Configurationally Stable C-N Axial Chirality

Summary: The first atroposelective synthesis of pyrrolo[3,4-b]pyridines catalyzed by N-heterocyclic carbene has been achieved, leading to a wide range of chiral atropisomers with high yields and excellent enantioselectivities (96-99% enantiomeric excess). Experimental results and density functional theory calculations demonstrate the high thermal stability of the C-N axial chirality in the product.

Photoredox Deaminative Alkylation in DNA-Encoded Library Synthesis

Copper-Mediated Cyclization of o-Hydroxyaryl Enaminones with 3-Indoleacetic Acids toward the Synthesis of 3-Indolmethyl- Chromones

Summary: A copper-mediated tandem decarboxylative coupling/annulation protocol was developed for the synthesis of 3-indolmethyl-chromones, with up to 97% yield. A one-pot method from o-hydroxy acetophenones, N, N-dimethylformamide dimethyl acetal, and 3-indoleacetic acids was also achieved. Derivatization of the products yielded various indolmethyl-substituted pyrimidines, some of which exhibited anti-influenza viral activities.
