The reaction was successfully scaled up to gram-scale by flow-electrocatalysis. Two key intermediates, rhodium(III) species, were isolated and fully characterized. Cyclovoltammetric analysis, X-ray photoelectron spectroscopy studies, and density functional theory calculations suggested the presence of a rhodium(III-IV-II-III) manifold.

Ru(II)- or Rh(III)-Catalyzed Annulation of Aromatic/Vinylic Acids withAlkylidenecyclopropanes via C-H Activation

Summary: An efficient and novel synthesis route for (E)-4-benzylideneisochroman-1-ones has been demonstrated using Ru(II) as a catalyst for the tandem cascade annulation of benzoic acids with alkylidenecyclopropanes. The reaction selectively delivers E-stereoselective 4-benzylideneisochroman-1-one derivatives in moderate to good yields, exhibiting completely different selectivity compared to previous methods. Furthermore, annulation with less-reactive beta C-H activation of vinylic acids with alkylidenecyclopropanes has been explored, leading to the synthesis of highly useful alpha-pyrone derivatives in the presence of an Rh(III) catalyst.

Rh(III)-Catalyzed Synthesis of Amino-isocoumarins with N-Functionalized Cyclic Carbonates via C-H/O-H Annulation

Summary: A practical method for accessing amino-isocoumarins through redox-neutral C-H/O-H annulation using a Rh(III) complex has been described. The use of N-functionalized cyclic carbonates is crucial for efficient catalytic turnover, leading to the preparation of a broad range of amino-isocoumarin derivatives with satisfactory yields. One derivative, an amino-isocoumarin conjugated with a selenocyano functionality, was found to be nearly four times as active as the marketed drug abiraterone against T47D cancer cells.

Scalable Electrochemical Aerobic Oxygenation of Indoles to Isatins without Electron Transfer Mediators by Merging with an Oxygen Reduction Reaction

The green protocol features the use of molecular oxygen as the sole oxidant and gram-scale preparation without an electron transfer mediator. A detailed mechanism of the reaction was elucidated using density functional theory calculations.

The strong coordinating ability of ortho- and meta-substituted pyridine boronates and pyrazoles as well as unsubstituted pyrimidine allows them to serve as the coupling partners. This method enables late-stage modification of the key precursor of roflumilast and compounds of pharmaceutical interest.

Electrooxidative Iridium-Catalyzed Regioselective Annulation of Benzoic Acids with Internal Alkynes

Rhodium-Catalyzed C4-Selective C-H Alkenylation of 2-Pyridones by Traceless Directing Group Strategy
