Palladium-Catalyzed Carbonylations: Application in Complex Natural Product Total Synthesis and Recent Developments

The selected examples include spinosyn A, callyspongiolide, perseanol, schizozygane alkaloids, cephanolides, and bisdehydroneostemoninine and related stemona alkaloids. The perspective also provides insights into recent advancements and future developments of palladium-catalyzed carbonylations.

Palladium-Catalyzed Chemodivergent Carbonylation of ortho-Bromoarylimine to Biisoindolinones and Spiroisoindolinones

Summary: A palladium-catalyzed carbonylative cyclization reaction of ortho-bromoarylimines is reported, allowing for the chemodivergent synthesis of functionalized biisoindolinones and spirocyclic isoindolinones. By switching the reaction temperatures and ligands, either product could be selectively obtained, and the biisoindolinone products could be easily achieved with catalyst loadings as low as 0.05 mol%. Further transformation of the biisoindolinone product is also described, providing a novel and concise approach to the biisoindoline diamine ligand.

Cascade Vinylation/8π-Electrocyclization and Cu(II)-Catalyzed Dehydrogenation toward Highly Stable Formally Antiaromatic Cycloheptatrienyl Anions

Summary: This study proposes a method for the synthesis of seven-membered systems through the chain elongation of nucleophilic propenes and subsequent electrocyclization reactions. The cascade reaction can yield either cycloheptadienes or bicycloheptenes, with the latter being formed via a reversible 6π-electrocyclization of an intermediate cycloheptadienyl anion. The electrocyclic nature of the ring-closing reactions is supported by computational calculations. Highly electron-deficient cycloheptatrienes can be obtained through oxidation reactions. Additionally, a retro-[2+2]-cycloaddition reaction was observed in a bicycloheptene derivative.

Summary: A highly enantioselective cascade carbonylation/annulation reaction has been established by Pd/chiral Lewis base relay catalysis, which assembles chiral quinolinones efficiently with excellent diastereo- and enantioselectivities from readily available starting materials under mild conditions. The palladium catalyst plays dual roles in this reaction, facilitating both the carbonylation process and the generation of the zwitterionic pi-allyl palladium intermediate.

Sustainable parts-per-million level catalysis with FeIII: One-pot cascade synthesis of 2,3-dihydroquinazolin-4(1H)-ones in water

Summary: A silica-supported iron complex has been identified as a highly active and reusable catalyst for the synthesis of medicinally important 2,3-dihydroquinazolin-4(1H)-ones. The catalyst showed high turnover number (TON) and turnover frequency (TOF), with successful synthesis of a diverse library of compounds in ppm levels of Fe using water as a solvent. The eco-friendly and sustainable protocol also demonstrated wide substrate scope, shorter reaction time, and gram-scale synthesis.
