Au(I)-Catalyzed Formal Intermolecular Carbene Insertion into Vinylic C(sp2)-H Bonds and Allylic C(sp3)-H Bonds

Xiao-Yuan Cui et al.

Summary: We report on a metal-catalyzed formal intermolecular carbene insertion reaction into the vinylic C(sp(2))-H bond of alkenes, with Au(I) catalyst proving to be the most efficient among the screened metal catalysts. This reaction provides a facile method to access C4-alkenyl isochromanones and isoquinolinones from corresponding diazo reagents and alpha-methylstyrenes, with moderate to good yields, and excellent chemoselectivity. Mechanistic studies suggest the existence of two possible reaction pathways: nucleophilic attack-selective elimination by the gold-carbenoid and cyclopropanation-ring-opening-olefin isomerization process. The structure of the diazo reagents has a significant impact, as diazo(thio)oxindoles react with alpha-methylstyrenes via formal allylic C(sp(3))-H insertion to afford 3-allyl(thio)oxindoles.

BF3•OEt2 catalyzed decarbonylative arylation/C-H functionalization of diazoamides with arylaldehydes: synthesis of substituted 3-aryloxindoles

Sengodagounder Muthusamy et al.

Summary: A metal-free BF3 center dot OEt2 catalyzed direct decarbonylative arylation of diazoamides with readily accessible aryl aldehydes under an open-air atmosphere was developed to afford 3-aryloxindoles via 1,2-aryl migration with high selectivity. The reaction offers an efficient pathway for 3-arylation of diazoamides under relatively mild conditions, which shows a high level of functional group tolerance of both electron-donating and electron-withdrawing groups with a broad substrate scope.

Visible-Light-Mediated Formal Carbene Insertion Reaction: Enantioselective Synthesis of 1,4-Dicarbonyl Compounds Containing All-Carbon Quaternary Stereocenter

Hua Zhang et al.

Summary: We have developed an efficient visible-light-mediated formal carbene insertion reaction using 1,3-diketones and diazoesters for the synthesis of enantioenriched 1,4-dicarbonyl compounds with a quaternary carbon center. By combining visible light and a Bronsted acid catalyst, we achieved chiral 1,4-dicarbonyl compounds in good yields with high enantioselectivities through a photochemical carbene transfer protocol.

Synthesis of Functionalized Indolobenzazepinones via Sc(OTf)3-Induced Ring Expansion/Annulation Reactions of 4-Diazoisoquinolin-3-ones with Isatins

Jianwei Xie et al.

Summary: In this study, an unexpected cascade ring expansion/annulation reaction was reported between 4-diazo-1,4-dihydroisoquinolin-3-ones and a variety of isatin derivatives in the presence of Sc(OTf)3 catalyst. Novel indolobenzazepinones were synthesized in good to excellent yields by excluding nitrogen and carbon dioxide gases. This method is characterized by high bond-forming efficiency, a novel reaction mechanism, readily available starting materials, commercially available cheap catalyst, broad substrate scope, and mild reaction conditions.

Facile and diastereoselective arylation of the privileged 1,4-dihydroisoquinolin-3(2H)-one scaffold

Dmitry Dar'in et al.

Summary: This study presents a practically convenient and streamlined protocol for introducing an aryl substituent at position 4 of the 1,4-DHIQ scaffold. The protocol involves Regitz diazo transfer and TfOH-promoted hydroarylation. Screening of the synthesized compounds against cancer cell lines confirmed high cytotoxicity of selected analogs, validating the potential of this new compound as an anticancer agent.

Chemical Research on Antitumor Isoquinoline Marine Natural Products and Related Compounds

Naoki Saito

Summary: This review summarizes the research on biologically active 1,2,3,4-tetrahydroisoquinoline-quinone (THIQ) family members isolated from Actinomycetes and marine organisms over the past five decades. Marine natural products and their reduced compounds have shown potential in antitumor profiles, with compounds like renieramycins and ecteinascidins attracting interest due to their unique structures and limited availability in nature.

A Selective C-C Bond Cleavage Strategy Promoted by Visible Light

Rafael D. C. Gallo et al.

Summary: The new reaction utilizing aryldiazoacetates and 1,3-diketones under visible light promotes C-C bond insertions to form 1,4-dicarbonyl compounds with high yields and selectivities. This straightforward and practical transformation is tolerant to air and moisture, without the need for metal catalysts. Mechanistic studies suggest the involvement of a key cyclopropanol intermediate resulting from an intramolecular rearrangement.

Catalytic Asymmetric Homologation of Ketones with α-Alkyl α-Diazo Esters

Fei Tan et al.

Summary: This study describes an enantioselective homologation of ketones with alpha-alkyl alpha-diazo esters using a chiral scandium(III) N,N'-dioxide catalyst to produce a series of optically active beta-keto esters with an all-carbon quaternary center. This method provides a highly selective and synthetically efficient strategy for the synthesis of enantioenriched compounds.
