Catalytic Atroposelective C7 Functionalisation of Indolines and Indoles

Saad Shaaban et al.

Summary: Axially chiral atropisomeric compounds, especially indole- and indoline-based frameworks, are important heterobiaryl classes widely applied in asymmetric catalysis and medicinal chemistry. The synthesis of C7-substituted indole biaryls and the analogous indoline derivatives is particularly challenging, and efficient methods for their synthesis are in high demand. Transition-metal catalysis is considered one of the most efficient methods for constructing atropisomers.

Bifunctional Ionic Liquid Catalyzed Multicomponent Arylsulfonation of Phenols with Aryl Triazenes and DABSO for the Synthesis of Diaryl Sulfones

Chengzong Tang et al.

Summary: The bifunctional Lewis acidic ionic liquid (LAIL) catalyzed multicomponent arylsulfonation reaction was developed, which transformed various aryl triazenes into aryl sulfonyl radicals by coupling with DABSO, and then coupled with phenoxy radicals to yield diaryl sulfones. This reaction showed good functional-group tolerance, gram-scale reactivity, and avoided the use of SO2 gas.

Copper-Catalyzed C7-Selective C-H/N-H Cross-Dehydrogenative Coupling of Indolines with Sulfoximines

Sonbidya Banerjee et al.

Summary: In this study, the Cu-catalyzed cross-dehydrogenative coupling of the C7 C-H bond of indolines with sulfoximines has been successfully achieved. The reaction can also be extended to the cross-dehydrogenative coupling of N-aryl 7-azaindoles with monoselectivity. The use of first-row copper catalysis, substrate scope, and late-stage synthetic applications are important practical features of this study.

From C4 to C7: Innovative Strategies for Site-Selective Functionalization of Indole C-H Bonds

Jian Wen et al.

Summary: The presence of hydrocarbons makes C-H functionalization an attractive alternative, with indole being an important heteroarene. Selective C-H functionalization at the benzene core of indoles is challenging, with efforts focusing on directing group development. Innovative strategies have been developed to selectively functionalize different positions of indoles, offering efficient methods for the synthesis of valuable substituted indoles.

Rh(III)-Catalyzed Selective C7 Halogenation of Indolines

Manisha et al.

Summary: An efficient Rh-catalyzed method has been developed for selective C7 halogenation of N-pyrimidyl indolines, producing corresponding halides in good to excellent yields. The strategy demonstrates broad substrate scope, excellent regioselectivity for C7 functionalization of indolines, and feasibility at gram scale level. Various control experiments were conducted to understand the reaction pathway, and the applicability of the methodology was demonstrated by synthesis of indoles and post-transformation of the C7 halogenated indolines into different valuable molecules.
