Bronsted Acid-Catalyzed Intramolecular 7-endo Hydroarylation Reaction of 1,5-Diaryl-1-pentynes

Summary: A Bronsted acid-catalyzed, transition metal-free intramolecular 7-endo hydroarylation reaction of 1,5-diaryl-1-pentynes has been developed. The use of 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP) as a solvent was found to be critical for this process. Using the present methodology, we have accomplished the synthesis of KGP-18, an analogue of the anticancer natural product combretastatin A4.

Transition-Metal-Catalyzed Denitrogenative Annulation to Access High-Valued N-Heterocycles

Summary: In recent years, the development of efficient methods to construct high-valued N-heterocyclic molecules has received significant attention due to their extensive applications in medicinal chemistry, drug discovery, and natural product synthesis. The transition-metal-catalyzed denitrogenative annulation of 1,2,3-triazoles and 1,2,3,4-tetrazoles has emerged as a powerful synthetic tool for the preparation of nitrogen-rich molecules. However, annulation of 1,2,3,4-tetrazole remains challenging. This Review summarizes the achievements and potential applications of denitrogenative annulation reactions in various fields.

Despite these advancements, the synthesis of chiral annulated products remains challenging. This review highlights recent advancements in asymmetric annulation reactions catalyzed by chiral CpxM(III) complexes, focusing on the mechanistic understanding of the developed protocols.

Summary: The construction of spiroisoindolinone indenes through a Bronsted acid-catalyzed reaction between iso-indolinone-derived propargylic alcohols and external aromatic nucleophiles is described. The reaction offers a modular approach for the synthesis of target compounds through an intercepted Meyer-Schuster rearrangement/intramolecular Friedel-Crafts alkylation relay, providing spiroindenes with good chemoselectivity and regioselectivity in moderate to high yields.

Summary: An efficient and atom-economic one-pot synthesis method for isoindolo[2,1-a]indol-6-ones has been developed, utilizing CuI/Pd(OAc)(2)-catalyzed intramolecular hydroamidation of alkynyl group and C-H dehydrogenative coupling of ortho-alkynyl-N-arylbenzamides. Oxygen is used as the oxidant and water is the sole by-product in this transformation. The reaction exhibits excellent tolerance towards various functional groups and provides isoindolo[2,1-a]indol-6-ones in good to high yields.

Bronsted Acid Promoted Sulfenylacyloxylation of Alkynes: Access to 4-Sulfenylisocoumarins

Summary: This study presents a electrophilic sulfenylacyloxylation reaction mediated by Bronsted acid (MsOH), in which alkynes are activated by in-situ formed sulfonium cations from bench-stable N-thioaryl/alkyl-succinimides, followed by intramolecular regioselective cyclization. This transformation enables the synthesis of structurally diverse 4-sulfenylisocoumarins in high yields (77-98%) with broad functional group compatibility. The reaction scalability and reusability of the phthalimide protecting group make this protocol synthetically viable.

This review comprehensively discusses the use of 2-nitrostyrene motifs in the synthesis of various N-heterocycles, highlighting their important features, reactivities, and mechanisms.
