Summary: An efficient and atom-economical palladium-catalyzed intramolecular cross dehydrogenative coupling (CDC) reaction has been developed for constructing highly pi-conjugated compounds using molecular oxygen as sole oxidant. The method has wide substrate scope, good functional group tolerance, and can generate fluorescence active compounds with high quantum yield. Additionally, the synthetic versatility of the method is showcased through Fe-catalyzed reductive isoxazole ring cleavage towards heteropolycyclic compounds.

Ruthenium(II)-Catalyzed C-C/C-N Coupling of 2-Arylquinazolinones with Vinylene Carbonate: Access to Fused Quinazolinones

Summary: This work presents a ruthenium(II)-catalyzed C-C/C-N annulation of 2-arylquinazolinones with vinylene carbonate to synthesize fused quinazolinones, showing excellent substrate tolerance and functional-group compatibility. The use of vinylene carbonate as an ethynol surrogate without external oxidants is highlighted, along with preliminary mechanistic studies and a proposed catalytic cycle.

Design, synthesis and biological activity of N-(amino)piperazine-containing benzothiazinones against Mycobacterium tuberculosis

Summary: A series of novel benzothiazinone derivatives containing a N-(amino)piperazine moiety, based on the structure of WAP-1902 discovered in the lab, were designed and synthesized as new anti-TB agents. Many compounds showed excellent in vitro activity against drug-sensitive MTB strain H37Rv and multidrug-resistant clinical isolates, with good safety index. Compound 1o, in particular, displayed low hERG cardiac toxicity and acceptable oral pharmacokinetic profiles, indicating its potential as a lead compound for future antitubercular drug discovery.

A microwave-assisted copper-mediated tandem approach for fused quinazoline derivatives

Summary: A method has been developed for the synthesis of fused-polycyclic systems via new C-N bond formation through a microwave-assisted copper-mediated oxidative coupling reaction of different aldehydes and quinazolines/benzimidazoles. The use of environmentally benign NH4OAc as the amine source in the presence of 2-propanol allows for a rapid and straightforward access to complex fused quinazoline derivatives.
