Oligodendrocyte progenitor development from the postnatal rat subventricular zone is regulated by the p75 neurotrophin receptor

Subhashini Joshi et al.

Summary: The precise timing of neural progenitor development and the correct balance between proliferation and differentiation are crucial to generating a functional brain. Lack of p75NTR reduced progenitor proliferation and caused premature oligodendrocyte differentiation and maturation both in vitro and in vivo, leading to aberrant early myelin formation. Our data reveal a novel role for p75NTR as a rheostat for oligodendrocyte production and maturation during myelin formation in the postnatal rat brain.

Neurotrophin signalling in the human nervous system

Sarah Ateaque et al.

Summary: This review article focuses on the relevance of neurotrophins and their tyrosine kinase receptors to the function and dysfunction in the human nervous system. It discusses the measurement of BDNF levels and presents recent findings on TrkB and TrkC signaling in human neurons from the authors' laboratory, including ligand selectivity and Trk activation by neurotrophins and non-neurotrophin ligands. The article also discusses ligand-induced down-regulation and re-activation of Trk receptors.

Remodelling of myelinated axons and oligodendrocyte differentiation is stimulated by environmental enrichment in the young adult brain

Madeline Nicholson et al.

Summary: This study found that environmental enrichment can induce structural remodeling of myelinated axons during young adulthood and globally increase the differentiation of oligodendrocyte progenitor cells and pre-myelinating oligodendroglia.

Plasticity in oligodendrocyte lineage progression: An OPC puzzle on our nerves

Georgina A. Craig et al.

Summary: Research has shown that myelin deposition in the central nervous system can be influenced by experience, with sensory enrichment increasing myelination and sensory or social deprivation decreasing myelination. Adaptive myelination is considered an essential component of new learning, driving nuanced alterations that ultimately lead to functional responses.

The effects of microglia- and astrocyte-derived factors on neurogenesis in health and disease

Tasuku Araki et al.

Summary: Hippocampal neurogenesis continues throughout life and is tightly controlled by environmental conditions, with microglia and astrocytes playing key roles in modulating the process. Glial cells release factors that support adult neurogenesis under normal conditions, but can also contribute to inflammation and hinder neurogenesis in diseases like Alzheimer's, depression, and epilepsy.

Chronic Administration of 7,8-DHF Lessens the Depression-like Behavior of Juvenile Mild Traumatic Brain Injury Treated Rats at Their Adult Age

Shih-Te Yang et al.

Summary: Traumatic brain injury (TBI) is a significant cause of mortality and morbidity among young people worldwide, with long-lasting sequelae including paralysis, epilepsy, and mental disorders like major depressive disorder. This study focused on the long-term effects of mild TBI in juvenile animals, revealing that even mild juvenile TBI treatment could lead to sustained adverse effects in adulthood, particularly in terms of depression-like behavior. The chronic administration of the TrkB agonist 7,8-DHF was found to lessen the mTBI-J treatment-induced depression-like behaviors in adult rats, suggesting its potential therapeutic use.

Identification of TrkB Binders from Complex Matrices Using a Magnetic Drug Screening Nanoplatform

Summary: The study demonstrates the effective immobilization of functional TrkB on magnetic iron oxide nanoclusters for drug discovery strategies targeting neurological disorders. The immobilized TrkB receptors can efficiently identify and bind compounds from mixtures, enabling rapid separation through magnetic isolation. The research suggests potential therapeutic effects of compounds interacting with TrkB found in Gotu Kola plant extract for dementia treatment.
