Protein tyrosine kinase inhibitor resistance in malignant tumors: molecular mechanisms and future perspective

Yang Yang et al.

Summary: Protein tyrosine kinases (PTKs) are a class of proteins that can phosphorylate tyrosine residues and their aberrant activation can lead to tumor growth. Tyrosine kinase inhibitors (TKIs) have been used in targeted therapy of cancer, but drug resistance limits their efficacy. Tumor microenvironment, cell death resistance, tumor metabolism, epigenetic modification, and abnormal drug metabolism are closely related to TKI resistance.

Receptor Tyrosine Kinase Signaling and Targeting in Glioblastoma Multiforme

Manali Tilak et al.

Summary: Glioblastoma multiforme (GBM) is a deadly cancer with limited response to existing therapies. Subtypes of GBM with distinct genetic signatures show aberrant activation of signal transduction pathways. Current research focuses on understanding these molecular alterations to develop more efficient targeted therapies.

Rabeprazole has efficacy per se and reduces resistance to temozolomide in glioma via EMT inhibition

Deepak Babu et al.

Summary: The study reveals a clinical association of the EMT-like process with glioma malignancy and poor survival, as well as the anticancer and temozolomide sensitizing effect of rabeprazole by repressing EMT. The increased expression of EMT-associated proteins in glioma biopsies may contribute to tumor malignancy and poor prognosis. Rabeprazole treatment attenuates glioma cell growth and migration, induces apoptosis, and sensitizes temozolomide resistance.

Mechanisms of temozolomide resistance in glioblastoma - a comprehensive review

Neha Singh et al.

Summary: Glioblastoma (GBM) is the most common primary malignant brain tumor in adults with a low median overall survival. Current standard treatment includes surgery followed by radiation and chemotherapy, with temozolomide (TMZ) being the first-choice chemotherapy drug. However, the development of resistance to TMZ is a major challenge in effective treatment, with multiple molecular mechanisms contributing to this resistance, such as DNA repair systems, signaling pathways, autophagy, epigenetic modifications, and microRNAs.
