Energetics, kinetics, and pathways of SNARE assembly in membrane fusion

Summary: The fusion process of transmitter-containing vesicles with plasma membranes plays a crucial role in neurotransmission and muscle contractions. This article reviews recent progress in understanding the assembly of synaptic SNARE proteins and the process of membrane fusion, highlighting the results obtained through single-molecule manipulation approaches and electric recordings of single fusion pores. The findings suggest that stepwise and cooperative SNARE assembly is responsible for stagewise membrane fusion.

Phosphorylation of VAMP8 was found to reduce spontaneous fusion under normal conditions while preassembling multiple lysosomes to increase fusion probability upon stimulation, impacting autophagosome maturation and chemotherapy drug resistance.
