Target nucleic acid amplification-free detection of Escherichia coli O157:H7 by CRISPR/Cas12a and hybridization chain reaction based on an evanescent wave fluorescence biosensor

Summary: A target nucleic acid amplification-free method called the CHANCE system was developed for rapid and sensitive detection of Escherichia coli O157:H7. The method showed a detection limit of 17.4 CFU/mL and could accurately detect E. coli O157:H7 within 50 minutes. It provides an ideal alternative for rapid and on-site detection of pathogenic bacteria in the environment and food.

Magnetic nanoparticles for food hazard factors sensing: synthesis, modification and application

This review summarizes the synthesis methods of MNPs and explores their modification with biological and abiotic functional groups, highlighting their recognition mechanisms. The advances and future perspectives of MNPs-based biosensors for FHFs detection are also discussed.

Cefepime-modified magnetic nanoparticles and enzymatic colorimetry for the detection of Listeria monocytogenes in lettuces

Summary: A novel sandwich assay was developed for the rapid separation and detection of pathogenic bacteria in food. It utilized antibiotic magnetic separation and enzymatic colorimetry. The method showed excellent bacterial capture capacity using PEG-mediated cefepime functionalized magnetic nanoparticles. The specificity for L. monocytogenes was ensured by using anti-L. monocytogenes monoclonal antibody as the second anchoring agent. The assay had a low limit of detection and could be completed in a short time.

Ultrasensitive quantification of pathogens in milliliters of beverage by filtration-based digital LAMP

Summary: To achieve rapid and ultrasensitive quantification of pathogens in large volume food sample, a filtration-based interfacial digital LAMP (idLAMP) system was developed. This system utilizes a nanoporous membrane for filtration and nanoporous hydrogel for digital amplification, allowing for digital counting of single bacteria at the membrane surface under nanoconfinement. The idLAMP successfully accomplished the quantitative detection of Escherichia coli in 100 mL water samples within 30 min, with a wide dynamic range from 0.09 to 900 cells/mL. This technique shows great potential in quantitative counting of single bacteria in large-volume food samples.

Potential reservoirs of antimicrobial resistance in livestock waste and treated wastewater that can be disseminated to agricultural land

Summary: In this study, the diversity and composition of microbial communities, antibiotic resistance genes (ARGs), virulence factor genes (VFGs), and nitrogen cycling genes in livestock manure, dairy lagoon effluent, and treated wastewater were examined using shotgun metagenomic sequencing. The presence of ARGs and VFGs in these sources pose a serious public health threat when applied to agricultural farmland.

Recent applications of rolling circle amplification in biosensors and DNA nanotechnology

Summary: Isothermal nucleic acid amplification techniques are crucial diagnostic tools for clinical applications and food safety inspections. Rolling circle amplification (RCA) is a highly efficient and robust method that meets the convenient amplification requirements for point-of-care testing. This review introduces the principles, species, and optimization strategies of RCA, and summarizes recent achievements in the design, development, and application of RCA biosensors. Additionally, the versatile application of RCA in DNA nanotechnology for bioimaging, drug delivery, and therapeutics is discussed. The current challenges and future perspectives of the RCA technique are also addressed.

Emerging insights into the use of carbon-based nanomaterials for the electrochemical detection of heavy metal ions

The review also highlights the role of carbon-based electrode materials in on-site detection and addresses potential challenges and future prospects.

Triplex PCR combined with magnetic separation strategy for rapid and specific detection of methicillin-resistant Staphylococcus aureus in hospital samples
