Inhibition of gasdermin D-dependent pyroptosis attenuates the progression of silica-induced pulmonary inflammation and fibrosis

Meiyue Song et al.

Summary: This study investigates the role of GSDMD-mediated pyroptosis in silicosis. The results demonstrate that GSDMD-dependent pyroptosis in macrophages is relevant to the progression of silicosis. Inhibiting this process could serve as a clinical strategy for mitigating silicosis.

Tauroursodeoxycholic acid (TUDCA) disparate pharmacological effects to lung tissue-resident memory T cells contribute to alleviated silicosis

Chao Li et al.

Summary: This study demonstrates the pharmacological effects of TUDCA on pulmonary inflammation and fibrosis induced by crystalline silica (CS). TUDCA inhibits the production of interferon (IFN)-gamma and interleukin (IL)-17A by pulmonary helper T (Th) cells, leading to a decrease in pathogenic TEM cells expansion. Furthermore, TUDCA's anti-fibrotic effects are associated with a decrease in IFN-gamma producing CD103- TEM-like and IL-17A producing CD103+ TRM-like T cells as well as restricting TRM-like Treg cells in the lung.

Gasdermin D pore structure reveals preferential release of mature interleukin-1

Shiyu Xia et al.

Summary: Inflammasomes activate caspase-1 to cleave GSDMD and generate mature cytokines like IL-1 beta and IL-18, which are released through pores formed by GSDMD. The GSDMD pore mediates IL-1 release by electrostatic filtering, favoring the passage of mature cytokines over their precursors.

Metformin inhibition of mitochondrial ATP and DNA synthesis abrogates NLRP3 inflammasome activation and pulmonary inflammation

Summary: The study found that metformin can attenuate COVID-19-induced ARDS by inhibiting NLRP3 inflammasome activation, IL-1 beta and IL-6 secretion, and by blocking ATP and mtDNA synthesis.

Inhibition of ER stress by targeting the IRE1α-TXNDC5 pathway alleviates crystalline silica-induced pulmonary fibrosis

Xi Chen et al.

Summary: Inhibition of ER stress alleviated crystalline silica-induced pulmonary fibrosis, with TXNDC5 playing a key role in fibroblast activation and being closely related to IRE1 alpha-XBP-1 signaling. Pharmacological inhibition of IRE1 alpha improved pulmonary function and delayed crystalline silica-induced lung fibrosis, demonstrating potential as novel therapeutic targets for silicosis.

Metabolic Adaptation of Macrophages as Mechanism of Defense against Crystalline Silica

Antonella Marrocco et al.

Summary: Silicosis is a lethal pneumoconiosis with no available therapy, affecting over 2.2 million people exposed to silica in the United States each year. Exposure to silica induces metabolic reprogramming of macrophages, impacting cell viability and cytokine production, which are crucial in the pathogenesis of silicosis. Mitochondrial and metabolic alterations induced by silica and LPS on macrophages are different, highlighting the importance of complex II activity and tricarboxylic acid cycle remodeling in macrophage survival and cytokine-mediated inflammation in silicosis.
