Age-associated B cells are heterogeneous and dynamic drivers of autoimmunity in mice

Summary: Age-associated B cells (ABCs) are a type of memory B cell expressing the transcription factor T-bet, formed under inflammatory conditions. In systemic lupus erythematosus (SLE), ABC frequency is correlated with disease and they are considered the source of autoantibody-secreting cells. The heterogeneity of ABCs and their relationship to other B cell subsets such as plasmablasts is unclear. The study shows that ABCs are dynamic and can be plasmablast precursors or undergo cycles of reactivation. Deletion of CD11c-expressing B cells reveals a direct role for ABC-like B cells in lupus pathogenesis.

Summary: Abnormal activation of the immune system is a known cause of lupus. Inhibiting the inflammasome and its downstream components has been shown to reduce disease severity in lupus mouse models. This study examines the effects of IL-1 beta loss on disease severity in a lupus-prone mouse model and identifies potential clinical relevance of TNF and IL-17 signaling pathways. Caution may be needed when inhibiting inflammasome or IL-1 beta signaling in SLE, especially in patients with renal involvement.

B-cell depletion with obinutuzumab for the treatment of proliferative lupus nephritis: a randomised, double-blind, placebo-controlled trial

Summary: This study compared the efficacy of obinutuzumab with standard therapies and placebo in the treatment of LN. The results showed that obinutuzumab achieved better complete renal responses at 52 and 104 weeks compared to standard therapies alone. It also showed improvements in other renal response measures, serologies, estimated glomerular filtration rate and proteinuria. Obinutuzumab was well tolerated and no new safety signals were identified.

The absence of certain guanine exchange factors in double-knock-out (DKO) mice leads to the accumulation of ABCs, which differentiate into pathogenic effector B cell populations with pro-inflammatory function. The upregulation of Tlr7 in DKO males further enhances the dissemination and pathogenicity of ABCs, resulting in severe pulmonary inflammation and early mortality.
