Summary: This study analyzed the DNA methylome and transcriptome of adult and in vitro shoot culture-induced rejuvenation cuttings of apple rootstock M9T337. The results showed a global decrease in DNA methylation during rejuvenation, which may be correlated with changes in the expression of DNA demethylase and DNA methyltransferase genes. In addition, a DNA methylation inhibitor was found to enhance adventitious rooting ability and promote the expression of adventitious rooting-related genes in apple rootstock.

Technologies and applications of single-cell DNA methylation sequencing

Summary: DNA methylation is a stable epigenetic modification occurring at CpG dinucleotides, playing essential roles in both physiological and pathological processes. In human diseases, especially cancer, aberrant DNA methylation has been observed. However, conventional methods for DNA methylation profiling require a large amount of DNA from heterogeneous cell populations, making it impractical for rare or circulating tumor cells. Therefore, the development of sequencing technologies that can accurately profile DNA methylation using small numbers of cells, or even single cells, is crucial. Excitingly, various single-cell DNA methylation sequencing and omics sequencing technologies have been developed, greatly advancing our understanding of the molecular mechanisms of DNA methylation. In this review, we summarize these methods, their applications in biomedical sciences, discuss technical challenges, and provide prospects for future research directions.

DNA Demethylation Induces Tree Peony Flowering with a Low Deformity Rate Compared to Gibberellin by Inducing PsFT Expression under Forcing Culture Conditions

Summary: This study found that compared to GA treatment, 5-azaC treatment can effectively induce tree peony flowering by rapidly inducing PsFT expression, reducing the methylation level in the PsFT promoter, and enhancing leaf photosynthetic efficiency, leading to a significant reduction in flower deformity rate. GO analysis revealed that differentially expressed genes related to methylation were mainly located in the thylakoid, associated with photosynthesis and response to stimulus, while genes closer to GA treatment had a wider distribution and were associated with various processes and regulations. Additionally, several candidate genes related to flowering regulation were identified, providing new ideas for improving tree peony forcing culture technology.
