Comparative Phenotyping of Two Commonly Used Chlamydomonas reinhardtii Background Strains: CC-1690 (21gr) and CC-5325 (The CLiP Mutant Library Background)

Ningning Zhang et al.

Summary: In this study, phenotypic comparisons were performed between two commonly used strains of Chlamydomonas, CC-1690 and CC-5325. The results showed that CC-5325 grew faster heterotrophically in the dark, but had lower photosynthetic efficiency and was more heat-sensitive than CC-1690. Additionally, CC-5325 had an intact cell wall with reduced thickness, and could not maintain a sustained circadian rhythm of phototaxis.

Systems-wide analysis revealed shared and unique responses to moderate and acute high temperatures in the green alga Chlamydomonas reinhardtii

Ningning Zhang et al.

Summary: Different intensities of high temperatures affect the growth of photosynthetic cells differently. This study investigated the responses of Chlamydomonas reinhardtii to 24-hour moderate (35°C) and acute (40°C) high temperatures. While both temperatures induced photoprotection, 35°C promoted growth and carbon uptake, while 40°C disrupted cell division and decreased photosynthetic efficiency. During recovery, DNA synthesis-related transcripts/proteins increased, while those involved in photosynthetic light reactions decreased.

Summary: This study compared the growth of two species of green algae under different culture conditions and found that moderate acetate concentrations enhance growth and biomass production. Light is required for optimizing growth rates and cell characteristics. Different growth parameters and biomass estimation methods can lead to varied responses to organic carbon. The variation in acetate concentration results in profound physiological acclimation in the algae and changes in cell characteristics under different culture conditions.

Histone H3K4 methyltransferases SDG25 and ATX1 maintain heat-stress gene expression during recovery in Arabidopsis

Ze-Ting Song et al.

Summary: This study investigated the role of histone H3K4 methyltransferases SDG25 and ATX1 in heat stress tolerance in Arabidopsis, demonstrating their importance in regulating stress-responsive gene expression during both heat stress and stress recovery stages. Mutations of SDG25 and ATX1 were found to decrease histone H3K4me3 levels, increase DNA cytosine methylation, and inhibit the expression of a subset of heat stress-responsive genes during stress recovery. ChIP-qPCR results confirmed the binding of ATX1 to chromatin associated with these target genes, highlighting the impact of histone H3K4me3 on DNA methylation and transcriptional memory in plants under heat stress.

Heat Shock Signaling in Land Plants: From Plasma Membrane Sensing to the Transcription of Small Heat Shock Proteins

Baptiste Bourgine et al.

Summary: Heat stress events limit crop productivity, leading plants to accumulate heat-shock proteins (HSPs) for thermal protection. HSP chaperones play a critical role in regulating protein folding and preventing misfolded protein formation, thereby avoiding heat-induced cell death. The HSP20 family is tightly repressed at low temperatures, suggesting a costly mechanism that can become detrimental under unnecessary conditions.

Chloroplast SRP43 autonomously protects chlorophyll biosynthesis proteins against heat shock

Shuiling Ji et al.

Summary: The chloroplast signal recognition particle 43 (cpSRP43) effectively protects several tetrapyrrole biosynthesis proteins from heat-induced aggregation, enhancing their stability during leaf greening and heat shock. The substrate-binding domain of cpSRP43 is sufficient for chaperoning LHCPs, while stabilization of tetrapyrrole biosynthesis clients requires the protein's chromodomain 2. cpSRP54 inhibits cpSRP43's chaperone activity towards tetrapyrrole biosynthesis proteins, but high temperature weakens their interaction, allowing cpSRP43 to serve as an autonomous chaperone for thermoprotection.
