Tracking Spatial Distribution Alterations of Multiple Endogenous Molecules during Lentil Germination by MALDI Mass Spectrometry Imaging

Yan-Xia Zhang et al.

Summary: Exploring the spatial distribution alterations of metabolites during lentil germination is crucial for understanding its nutritional value, physiological function, and metabolic pathway. This study established an effective MALDI-MSI method to visualize the changes in spatial localization of 53 metabolites in lentils during germination. The results showed specific distribution patterns of different metabolites, with certain lipids mainly located in the cotyledons and others spreading throughout the lentil tissue during early germination. Heat map analysis demonstrated the correlation between lipid content changes and germination time, supporting the potential use of germinated lentils as nutraceutical or functional food.

MALDI Imaging Assisted Discovery of a Di-O-glycosyltransferase from Platycodon grandiflorum Root

Weiwei Tang et al.

Summary: A matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI MSI) assisted genome mining strategy was applied to discover a glycosyltransferase (GT) named PgGT1 from the root of Platycodon grandiflorum. PgGT1 was found to catalyze the synthesis of platycoside E (PE) by sequentially attaching two beta-1,6-linked glucosyl residues to the glucosyl residue at the C3 position of platycodin D (PD). The study also revealed the crucial roles of residues S273, E274, and H350 in stabilizing the sugar donor and positioning the glucose for optimal glycosylation. These findings provide insights into the biosynthetic pathway of PE and have potential implications in industrial biotransformation.

Summary: This study analyzed the spatial distribution of diterpenoids in Salvia miltiorrhiza and S. grandifolia using metabolomics and mass spectrometry imaging techniques. The results showed that S. miltiorrhiza contained mainly abietane-type norditerpenoid quinones with specific distribution in the roots, while S. grandifolia contained mainly phenolic abietane-type tricyclic diterpenoids with wider distribution in the roots. Additionally, the leaves of S. grandifolia were rich in tanshinone biosynthesis precursors, while those of S. miltiorrhiza were rich in phenolic acids. Genes involved in tanshinone biosynthesis were differentially expressed in the two species' roots. Summary: Nod factors are important signaling molecules produced by rhizobia in response to flavonoids for root nodule formation in legumes. It is hypothesized that they could also enhance yield and positively affect the growth of non-legumes. To evaluate this, rapeseed treated with Nod factor-based biofertilizers were studied, and it was found that the biofertilizers increased the concentration of lignin, hemicellulose, pectin, and cellulose in the stems. Additionally, the concentration of certain quercetin and kaempferol derivatives increased, while the concentration of isorhamnetin dihexoside decreased. The increase in structural components and flavonoids in the stems may improve lodging resistance, fungal infection resistance, and resistance to herbivorous insects.

Summary: DESI-MSI is a powerful tool for analyzing the distribution of metabolites in biological tissues, and cryosectioning using transparent adhesive films can improve the analysis of metabolites in strawberry fruits and other plant tissues. The study demonstrated that adhesive film-prepared cryosections maintained original structures, allowing for the visualization of amino acids, sugars, organic acids, and flavonoids in the tissues.

Phosphorus deprivation affects composition and spatial distribution of membrane lipids in legume nodules

Dhiraj Dokwal et al.

Summary: Phosphorus (P) is an essential macronutrient for symbiotic nitrogen fixation (SNF) in legumes, and its deficiency can impair SNF activity and affect the accumulation of phosphate in roots and nodules. Under P deficiency conditions, membrane lipids in nodules exhibit changes, with alterations in the distribution of phospholipids (PL) species. Specific phosphatidylcholine (PC) species in nodules may play different roles in response to P stress, leading to heterogeneous distribution within nodules.

The GORKY glycoalkaloid transporter is indispensable for preventing tomato bitterness

Yana Kazachkova et al.

Summary: By studying nine bitter-tasting accessions, including the 'high tomatine' Peruvian landraces, researchers found a deletion in the GORKY gene, which mediates the subcellular localization of alpha-tomatine during fruit ripening; GORKY exports alpha-tomatine and its derivatives from the vacuole to the cytosol, facilitating the conversion of the entire alpha-tomatine pool to non-bitter forms; these findings suggest that GORKY activity is a notable innovation in the process of tomato fruit domestication and breeding.
