IGF2BP1-regulated expression of ERRα is involved in metabolic reprogramming of chemotherapy resistant osteosarcoma cells

Summary: Doxorubicin (Dox) is the standard treatment for osteosarcoma (OS), but drug resistance limits its effectiveness. This study investigated metabolic reprogramming and its regulation in Dox-resistant OS cells. The results showed increased ATP levels and altered metabolic profiles in Dox-resistant cells, which was associated with elevated expression of estrogen-related receptor alpha (ERR alpha). Mechanistic analysis revealed increased mRNA stability in chemoresistant cells, possibly due to methylation of the 3' untranslated region of ERR alpha mRNA by N-6-methyladenine, leading to recruitment of IGF2BP1 and increased mRNA stability. Knockdown of IGF2BP1 reversed the metabolic alterations and downregulated ERR alpha, indicating the oncogenic role of the IGF2BP1/ERR alpha axis in Dox-resistant OS cells. Clinical analysis also showed correlation between IGF2BP1 and ERR alpha expression with OS progression.
