The COPS3-FOXO3 positive feedback loop regulates autophagy to promote cisplatin resistance in osteosarcoma

Summary: This study found that COPS3 is significantly upregulated in osteosarcoma tissues with poor response to preoperative chemotherapy, and its inhibition can make tumor cells more sensitive to cisplatin treatment. Further mechanistic investigations revealed that COPS3 promotes cisplatin-induced autophagy and maintains a high level of autophagy by regulating the stability of FOXO3. Therefore, COPS3-mediated autophagy inhibition may be a promising therapeutic strategy for overcoming cisplatin resistance in osteosarcoma.

Additionally, the usage of distal polyadenylation site (PAS) in the 3'UTR was reduced under DDP treatment, leading to a further decrease in the expression of the novel XBP1 transcript. Moreover, we found that XBP1 transcripts with shorter 3'UTR were more stable and enhanced DDP resistance.
