Macrophage-evading and tumor-specific apoptosis inducing nanoparticles for targeted cancer therapy

Summary: A biomimetic nanosystem was developed to escape the mononuclear phagocyte system (MPS) and target tumor tissues. The nanoparticles were coated with fibroblast cell membrane expressing tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), which reduced macrophage endocytosis and increased tumor cell uptake. Additionally, encapsulated chloroquine (CQ) suppressed macrophage uptake and synergized with TRAIL to induce tumor cell apoptosis.

Summary: Concerns about the efficacy of pegylated drugs arise due to the presence of antibodies against polyethylene glycol (PEG) in healthy individuals. This study evaluated the prevalence of anti-PEG antibodies in acute lymphoblastic leukemia (ALL) patients before and/or after the first administration of pegylated E.coli asparaginase (PEG-ASNase). The results showed that pre-existing anti-PEG antibodies were detected in a considerable proportion of patients with ALL and although they did not inhibit PEG-ASNase activity, they were associated with lower serum PEG-ASNase activity levels. Patients with pre-existing antibodies may experience mild to moderate hypersensitivity reactions after their first administration of PEG-ASNase, which can be successfully addressed by re-challenge.

Anti-PEG antibodies compromise the integrity of PEGylated lipid-based nanoparticles via complement

Summary: In this study, the consequences and mechanisms of complement activation by anti-PEG antibodies interacting with various types of PEGylated lipid-based nanoparticles were investigated. The activation of complement by anti-PEG antibodies can compromise the integrity of nanoparticles, leading to premature drug release or exposure to serum proteins. The levels of pre-existing anti-PEG IgM antibodies in plasma of healthy donors correlated with the degree of complement activation induced upon exposure to PEGylated liposomes and mRNA LNPs.

The long-circulating effect of pegylated nanoparticles revisited via simultaneous monitoring of both the drug payloads and nanocarriers

Summary: The study revisits the long-circulating effect of mPEG-PCL nanoparticles by monitoring the movement of drug payloads and nanocarriers. It demonstrates that pegylation can extend the circulation time of nanocarriers in the blood, although nanocarriers with dense PEG decoration are cleared quickly, and DOX is cleared faster than the nanocarriers. The study also shows that hepatic accumulation is the highest among all organs investigated but is inversely proportional to blood circulation time, suggesting possible leakage of DOX from the nanocarriers. Reduction in particle size and pegylation prove to be effective in extending circulation time and reducing uptake by organs of the mononuclear phagocytic system.
