Hybrid-Membrane-Decorated Prussian Blue for Effective Cancer Immunotherapy via Tumor-Associated Macrophages Polarization and Hypoxia Relief

Lin Hou et al.

Summary: This study successfully addresses the limitations of tumor-associated macrophages and hypoxia conditions in cancer immunotherapy by developing a hollow mesoporous Prussian blue nanosystem with mannose decoration and hydroxychloroquine adsorption. In vivo experiments demonstrate that this system effectively inhibits tumor growth, induces macrophage polarization, promotes cytotoxic T lymphocytes infiltration, and alleviates hypoxia microenvironment.

Impact of Superparamagnetic Iron Oxide Nanoparticles on THP-1 Monocytes and Monocyte-Derived Macrophages

Christina Polasky et al.

Summary: In this study, the impact of MPI optimized dextran-coated SPIONs on monocytic cells was investigated. It was found that SPIONs were rapidly taken up by monocytes and did not affect cell viability. However, SPION uptake hindered monocyte differentiation into macrophages and led to changes in cytokine secretion.

Iron oxide nanoparticles size-dependently activate mouse primary macrophages via oxidative stress and endoplasmic reticulum stress

Haoran Ying et al.

Summary: This study investigated the impact of iron oxide nanoparticles (IONPs) on immune responses in mice and macrophages. The results showed that IONPs could induce inflammation, and the effects were size dependent. Analysis also revealed that larger particles activated immune responses, endoplasmic reticulum stress, and oxidative stress. Additionally, blocking actin polymerization, endoplasmic reticulum stress, or oxidative stress could attenuate the inflammatory response caused by IONPs.

Engineering manganese ferrite shell on iron oxide nanoparticles for enhanced T1 magnetic resonance imaging

Muyao Li et al.

Summary: In this study, we successfully improved the T-1 contrast of iron oxide (IO) nanoparticle by controlling the thickness and designed an effective high-performance T-1 contrast agent. Doping Mn (II) ions into the shell of IO nanoparticle (IO@MnIO) significantly enhanced the T-1 relaxivity, especially at a thickness of -0.8 nm. In addition, by optimizing the interfacial structure, we successfully reduced the T-2 enhancing effect caused by doped Mn (II) ions, limiting the interference of increased T-2 relaxivity on T-1 contrast imaging.

Surface carboxylation of iron oxide nanoparticles brings reduced macrophage inflammatory response through inhibiting macrophage autophagy

Di Deng et al.

Summary: This study synthesized carboxyl-modified SPIO nanoparticles and found that compared with unmodified nanoparticles, carboxyl-modified particles reduced autophagy response in macrophages, leading to a milder inflammatory response. The increased amount of albumin adsorption on the surface of carboxylated nanoparticles was identified as a potential reason for the reduction in autophagy and inflammation.

The multifaceted roles of macrophages in bone regeneration: A story of polarization, activation and time

Claudia Schlundt et al.

Summary: Macrophages are a highly plastic and multifunctional cell population found in all tissues of the human body. In bone, different subpopulations of macrophages play essential roles in the cascade of regeneration and reconstitution, participating in both the inflammatory phase and later phases of healing. The M1/M2 paradigm may not fully capture the dynamic plasticity of macrophages, and understanding the specific mechanisms driving macrophage function in impaired healing scenarios could lead to new therapeutic targets.
