Low birth weight and small for gestational age are associated with complications of childhood and adolescence obesity: Systematic review and meta-analysis

Summary: Recent studies have shown that being born with low birth weight (LBW) or being small for gestational age (SGA) is associated with an increased risk of insulin resistance and type 2 diabetes (T2D) in children and adolescents. Children and adolescents born as SGA have a 2.33-fold higher risk of developing T2D compared to those born with adequate size for gestational age. These findings suggest the importance of screening tests and early preventive strategies for this potentially vulnerable subgroup.

Obesity-Associated GNAS Mutations and the Melanocortin Pathway

Summary: Variants in the GNAS gene have been linked to childhood obesity, showing the importance of genetic factors in this condition. Mutations in GNAS affect GPCR signaling pathways, leading to a wide range of clinical manifestations. Early diagnosis through genetic screening and targeted treatment with melanocortin agonists may improve outcomes for children with severe obesity.

Summary: Pseudohypoparathyroidism is a post-receptorial hormone resistance disorder caused by a partial deficiency of the alpha subunit of the stimulatory G protein. In addition to revealing the molecular basis of PHP, different subtypes and diseases associated with genetic alterations in other genes of the PTH/PTHrP pathway have been identified. The clinical and molecular overlap among different diseases makes both differential diagnosis and genetic counseling challenging.
