Determining critical overlap concentration of polyethylene oxide to support excipient safety assessment of opioid products

Summary: The intravenous administration of abuse-deterrent opioid products, containing high molecular weight polymeric excipients like polyethylene oxide (PEO), carries high safety risks due to previous in vivo studies showing adverse responses such as intravenous hemolysis and kidney damage. This study focuses on determining the critical overlap concentration (c*) at various PEO molecular weights to explain differences in observed adverse responses, suggesting that the viscoelastic regime of the polymer solution and localized polymer concentration above c* may contribute to adverse in vivo responses.
