Cisplatin toxicity in the developing brain displays an absolute requirement for caspase-3

Kelvin K. Hui et al.

Summary: Cisplatin-induced cell death in the developing CNS is highly dependent on caspase-3, and a delayed form of necroptosis-like cell death occurs. These findings suggest a coordinated regulation of genotoxic stress response in vivo.

7-hydroxycoumarin modulates Nrf2/HO-1 and microRNA-34a/SIRT1 signaling and prevents cisplatin-induced oxidative stress, inflammation, and kidney injury in rats

Demiana H. Sami et al.

Summary: In this study, the protective effect of the antioxidant 7-hydroxycoumarin (7-HC) against cisplatin-induced acute kidney injury was investigated. Results showed that 7-HC prevented kidney dysfunction and tissue injury, suppressed oxidative stress and inflammation, and modulated several signaling pathways. Molecular docking revealed the potential binding of 7-HC to specific proteins involved in these pathways.

Regulation of Keap-1/Nrf2/AKT and iNOS/NF-κB/TLR4 signals by apocynin abrogated methotrexate-induced testicular toxicity: Mechanistic insights and computational pharmacological analysis

Ahmed M. Sayed et al.

Summary: This study demonstrated the protective effects of apocynin (APO) against testicular damage induced by methotrexate (MTX), showing significant benefits such as increased hormone levels, restored tissue structure, and suppressed inflammation. APO exerted its protective effects through mechanisms including enhancing antioxidant status and reducing inflammation responses. Further investigation into the clinical application of APO in MTX-treated patients is warranted based on these promising findings.

Trans-ferulic acid ameliorates cisplatin-induced testicular damage via suppression of TLR4, P38-MAPK, and ERK1/2 signaling pathways

Emad H. M. Hassanein et al.

Summary: The study demonstrated that TFA has potential protective effects against CDDP-induced testicular injury by reducing oxidative stress and inflammatory response. It also enhanced the cytotoxic effect of CDDP in vitro through induction of apoptosis in both early and late stages. Additionally, TFA modulated various signaling pathways related to inflammation and cytotoxic activity.

Roflumilast protects from cisplatin-induced testicular toxicity in male rats and enhances its cytotoxicity in prostate cancer cell line. Role of NF-κB-p65, cAMP/PKA and Nrf2/HO-1, NQO1 signaling

Basel A. Abdel-Wahab et al.

Summary: The study found that ROF can reverse CIS-induced abnormalities in sperm characteristics, normalize serum testosterone level, and improve testicular and epidydimal weights, as well as restore normal testicular structure. ROF also increases intracellular cAMP level, PKA and HO-1 activities, Nrf2, NQO-1 and HO-1 gene expression, improves testicular oxidative stress parameters and inflammatory mediators, and reduces proapoptotic proteins' expression. In addition, ROF enhances the anticancer efficacy of CIS, induces Nrf2, HO-1, NQO-1 gene expression, inhibits NF-kappa beta p65 gene expression induced by CIS, and enhances its apoptotic effect in PC3 cells.

Dimethyl fumarate improves cognitive deficits in chronic cerebral hypoperfusion rats by alleviating inflammation, oxidative stress, and ferroptosis via NRF2/ARE/NF-kappa B signal pathway

Nao Yan et al.

Summary: The study demonstrates the neuroprotective effect of DMF in improving cognitive deficits and reversing hippocampus neuronal damage and loss caused by CCH. DMF reduces inflammation, oxidative stress, and inhibits ferroptosis in hippocampal neurons, thus improving cognitive impairment. The upregulation of the NRF2/ARE signaling pathway and related gene expressions contribute to the neuroprotective role of DMF.
