Integrated analysis of transcriptome and metabolome data reveals insights for molecular mechanisms in overwintering Tibetan frogs, Nanorana parkeri

Summary: In this study, a comprehensive transcriptomic and metabolomic analysis of N. parkeri was conducted, revealing altered energy metabolism and gene expression in overwintering frogs compared to summer-active ones. Up-regulated genes mainly participated in signal transduction and immune responses, while down-regulated genes were mainly involved in metabolic processes. Additionally, differentially expressed metabolites were mainly involved in amino acid biosynthesis and metabolism. These findings enhance our understanding of winter survival mechanisms in amphibians.

Summary: Hibernation reduces the barrier function of colonic mucosa in Daurian ground squirrels, leading to a decrease in the thickness and depth of the mucosa, a reduction in the number of goblet cells, and damage to microvilli structure. The concentrations of proteins and molecules, as well as the enzymatic activity of DAO, are increased in the serum and colon. Additionally, inflammation, apoptosis-anti-apoptosis, and FXR-SHP signaling may be involved in the changes in intestinal gut permeability during the torpor-arousal cycle.

The Protective Effects on Ischemia-Reperfusion Injury Mechanisms of the Thoracic Aorta in Daurian Ground Squirrels (Spermophilus dauricus) over the Torpor-Arousal Cycle of Hibernation

Summary: This study investigated the protective mechanisms involved in vascular ischemia-reperfusion injury during the torpor-arousal cycle in Daurian ground squirrels. The results showed that during the interbout-arousal period, there was a decrease in oxidative stress and pro-inflammatory factors, as well as an increase in anti-inflammatory factors, which induced controlled phenotypic switching of thoracic aortic smooth muscle cells and restoration of endothelial permeability.

Summary: Hibernating animals exhibit intrinsic cardiac protection against ischemia-reperfusion injury and cardiac arrhythmias in winter, which involves upregulation of transcriptional factors, anti-apoptotic factors, nitric oxide synthase, and antioxidant enzymes, as well as reduction in CaMKII oxidation level. The resistance to arrhythmias and sudden cardiac death is associated with upregulation of catalase and downregulation of CaMKII activation. Understanding these cardiac adaptive mechanisms may provide new strategies for protecting non-hibernating animals, including humans, from I/R injury and fatal arrhythmias.
