In silico screening and in vitro validation of phytocompounds as multidrug efflux pump inhibitor against E. coli

Summary: Multiple drug resistance (MDR) in bacteria has become a global issue, leading to reduced efficacy of antibiotics and increased therapeutic failure. This study aimed to identify potential inhibitors for the efflux pump of AcrB protein in E. coli using molecular docking. Among the 19 screened phytocompounds, chlorogenic acid showed the highest binding affinity and may serve as a promising efflux pump inhibitor in combination therapy against E. coli.

Inhibitory effect of baicalein against glycation in HSA: an in vitro approach

Summary: Hyperglycaemia accelerates aging significantly. Inhibiting glycation can alleviate diabetes problems. Human serum albumin was studied as a model protein to investigate glycation and antiglycation mediated by methyl glyoxal and baicalein. Glycation of Human Serum Albumin (MGO-HSA) induced by a Methylglyoxal (MGO) incubation period resulted in hyperchromicity, decreased tryptophan and intrinsic fluorescence, increased AGE-specific fluorescence, and reduced mobility. Structural and functional changes in glycated HSA, such as carbonyl groups on ketoamine moieties, are associated with physiological problems including diabetes mellitus and cardiovascular disease.

In silico and POM analysis for potential antimicrobial agents of thymidine analogs by using molecular docking, molecular dynamics and ADMET profiling

Summary: In this study, five 5'-O-(myristoyl)thymidine esters were synthesized and their chemical structures were determined by physicochemical, elemental, and spectroscopic analyses. In vitro antimicrobial tests and molecular docking studies showed promising antibacterial activity of these ester analogs. Molecular dynamics simulations and pharmacokinetic predictions further confirmed their stability and potential applications. SAR investigation revealed that myristoyl (C-14) and lauroyl (C-12) acyl chains combined with deoxyribose exhibited the best activity against bacterial and fungal pathogens.

Summary: Resistance to aminoglycosides in Pseudomonas aeruginosa remained constant over 2 decades, indicating the effectiveness of antibiotic stewardship programs. Clinically relevant AME genes were found in 14% of isolates, and mutations in mexZ and armZ genes were common. These findings highlight the ongoing problem of aminoglycoside resistance and suggest potential targets for novel therapeutics.

Summary: In the past two decades, antimicrobial resistance (AMR) has exponentially increased, posing a serious threat to human health. AMR is a leading cause of mortality worldwide. Despite a successful discovery of new antibiotics in the 20th century, little progress has been made in this area recently. Due to the rise in AMR and the slow progress in antibiotic discovery, there is an urgent need for new intervention methods to combat infectious diseases. One approach is to search for biofilm and quorum sensing inhibitors, and plants are a rich source of compounds that could exhibit these properties.

Summary: Insight gained into the mechanistic binding of bovine serum albumin with doxofylline revealed a spontaneous and exothermic process, along with pronounced structural changes in BSA upon binding with doxofylline. The study also explored the influence of metal ions and vitamins on the binding affinity of the BSA-doxofylline system. Additionally, both in vitro and in silico analyses supported the strong interaction between BSA and doxofylline, suggesting its potential for optimal distribution into the bloodstream for asthma treatment.

Plumbagin inhibits quorum sensing-regulated virulence and biofilms of Gram-negative bacteria: in vitro and in silico investigations

Computational studies provided insights into the possible modes of action, showing that the protein complexes remained stable under physiological conditions.
