Investigation of the interactions of HSA and SARS-CoV-2 papain-like protease against eugenol for novel COVID-19 drug discovery: spectroscopic and insilico study

High-throughput virtual screening approach of natural compounds as target inhibitors of plasmepsin-II

Summary: In this study, structure-based virtual screening methods were used to identify potential plasmepsin II inhibitors for the development of antimalarial drugs. A total of 58 quinoline derivatives were screened using molecular docking, ADMET analysis, molecular dynamics simulation, and binding free energy calculations. The top-21 compounds with the highest binding affinity were further evaluated for their pharmacokinetic properties and toxicity using ADMET analysis. Two potential inhibitors, M49 and M53, were identified and their stability was confirmed by molecular dynamics simulation. This study provides insights for the prediction and design of novel antimalarial drugs.

Molecular modeling study of pyrrolidine derivatives as novel myeloid cell leukemia-1 inhibitors through combined 3D-QSAR, molecular docking, ADME/Tox and MD simulation techniques

Summary: This study used pyrrolidine derivatives to investigate the structural requirements for designing novel Mcl-1 inhibitors. Three models (CoMSIA, CoMFA, and HQSAR) were created using QSAR molecular modeling techniques, and the results showed good stability and predictability. Four new compounds with stronger inhibitory activity were designed based on the obtained results. ADME/Tox results and pharmacokinetic properties suggested that these compounds are orally bioavailable and have good permeability.

A new compound with comparable activity to darunavir was identified and evaluated. The study suggests that this new molecule could be further investigated and used as a potential inhibitor of HIV-1 protease, and the method used in this study could be applied to search and design new inhibitors.

The physicochemical and ADMET/pharmacokinetic properties of the proposed molecules suggested their bioavailability and good gastrointestinal absorption.
