Recent development of multi-targeted inhibitors of human topoisomerase II enzyme as potent cancer therapeutics

Vishal Singh et al.

Summary: Multi-target therapies are a viable option to overcome drug-resistant cancer cells. With the use of a single drug against multiple structurally similar sites, it modulates several vital cellular pathways and reduces the usage of multiple drugs. However, drug resistance against anti-cancer agents targeting topoisomerase II has been observed, leading to the proposal of multi-target therapies to overcome different resistance mechanisms. This review discusses the binding cavity characteristics for drug interactions, potent inhibitors, and drug resistance mechanisms in various types of cancers.

Molecular docking and molecular dynamics simulation study on the toxicity mechanism of bongkrekic acid

Hongmei Li et al.

Summary: In this study, MD simulations were used to investigate the toxic mechanism of BKA on ANT protein translocation. The results showed that BKA mainly blocked ANT protein translocation through Van der Waals force, hydrophobic and hydrogen bonding interactions. Strong interactions with residues Gln-93, Tyr-196, Arg-287, and Arg-245 were found to affect the conformational changes of ANT.

Modulating mitofusins to control mitochondrial function and signaling

Emmanouil Zacharioudakis et al.

Summary: Mitofusins are proteins that regulate mitochondrial fusion, a process with significant impact on cellular processes. This study identifies small molecules that can activate or inhibit Mitofusins' activity, thereby modulating mitochondrial fusion and functionality. Inhibition of mitochondrial fusion leads to certain physiological changes such as caspase-3/7 activation and DNA damage. The findings highlight the importance of Mitofusins' conformational changes and oligomerization in enabling mitochondrial fusion. The study provides insights into the function and regulation of Mitofusins and offers potential small molecules for pharmacological targeting.
