Summary: COVID-19 has caused significant damage globally, and targeting the main protease (Mpro) of SARS-CoV-2 with existing drugs shows promise. Through molecular docking and virtual screening, glecaprevir and nelfinavir are identified as potentially effective antiviral drugs against Mpro. Other FDA-approved drugs also show potential and further research is needed.

Inhibitors of Heptosyltransferase I to prevent heptose transfer against antibiotic resistance of E. coli: Energetics and stability analysis by DFT and molecular dynamics

Summary: In this study, virtual screening was used to identify potential hit molecules from the ZINC, PubChem, and CHEMBL databases that can inhibit HEP I and reduce antibiotic resistance in E. coli. Pharmacokinetics and molecular dynamics simulations were performed to evaluate the bioavailability and conformational stability of the selected hits. The results suggest that three candidates, hit 1 (CHEMBL1438912), hit 3 (CHEMBL13895), and hit 4 (CHEMBL320880), show promising inhibitory potential and could be further investigated for antibacterial drug development.

Inhibitors of Plasmepsin X Plasmodium falciparum: Structure-based pharmacophore generation and molecular dynamics simulation

Summary: Malaria is a life-threatening disease caused by Plasmodium parasites and has posed a significant health challenge globally due to antimalarial resistance. Discovering effective new antimalarial medicines is crucial to combat resistance to current therapies. In this study, a protease called Plasmepsin X (PMX) was identified as playing a key role in the invasion of parasites, leading to the identification of potential inhibitors through structure-based pharmacophore generation and virtual screening. These new inhibitors show promise for further in vivo studies as antimalarial drugs.
