Structure of the membrane-bound formate hydrogenlyase complex from Escherichia coli

Summary: The membrane-bound formate hydrogenlyase (FHL) complex is a prototypical hydrogen-producing enzyme that connects formate oxidation to proton reduction. It has attracted intense interest due to its efficient hydrogen production during fermentation, reversibility, and evolutionary link to respiratory complex I. Despite being studied for over a century, the atomic structure of FHL remained unknown. In this study, cryo-EM structures of FHL were determined, revealing the importance of conserved loops and a metal-binding site near the interface of FdhF and HycF subunits.
