Phase separation of SGS3 drives siRNA body formation and promotes endogenous gene silencing

Huijuan Tan et al.

Summary: The formation of siRNA bodies is essential for siRNA production and gene silencing. SGS3 form condensates via phase separation and interact with RDR6 to drive the assembly of siRNA bodies in cytoplasm. Dysfunction in translation and mRNA decay can increase the number of siRNA bodies, while DCL2/4 mutations enhance their size. Purification of SGS3 condensates reveals the presence of numerous RNA-binding proteins and siRNA processing components.

Innate, translation-dependent silencing of an invasive transposon in Arabidopsis

Stefan Oberlin et al.

Summary: The co-evolution between hosts' and parasites' genomes shapes diverse pathways of acquired immunity based on silencing small (s)RNAs. In plants, the translation of short subgenomic mRNA (shGAG) during ribosomal stalling triggers a translation-dependent silencing (TdS) response that serves as a primary defense against de novo invasions.

Identification of a Transferrable Terminator Element That Inhibits Small RNA Production and Improves Transgene Expression Levels

Felipe Fenselau de Felippes et al.

Summary: The role of terminators in gene expression regulation and transcription termination in plants is explored in this study. The researchers identified a key element in the Arabidopsis HSP18.2 terminator that is essential for high level transgene expression. This sequence improves transcription termination, reduces read-through events, and decreases small RNA production. Additionally, the identified sequence also enhances splicing efficiency and promotes gene expression. The findings provide valuable insights into the molecular mechanisms of plant terminators and offer possibilities for manipulating gene expression through the manipulation of 3 ' regulatory regions.

Reprogramming of RNA silencing triggered by cucumber mosaic virus infection in Arabidopsis

Maria Luz Annacondia et al.

Summary: RNA silencing plays a crucial role in mediating virus resistance in plants. The interaction between viruses and RNA silencing machinery is complex, involving the loading of viral small interfering RNAs (vsiRNAs) into host ARGONAUTE (AGO) proteins. This study shows that vsiRNAs from cucumber mosaic virus (CMV) can affect endogenous sRNA levels and displace other endogenous sRNAs when loaded into AGO proteins. Additionally, the viral suppressor of RNA silencing (VSR) 2b counteracts this by loading vsiRNAs and mRNA-derived siRNAs to affect gene expression.

Ribosome stalling and SGS3 phase separation prime the epigenetic silencing of transposons

Eun Yu Kim et al.

Summary: This study demonstrates that plant transposon RNAs undergo frequent ribosome stalling during translation, leading to RNA truncation and localization to cytoplasmic siRNA bodies, which are essential for RDR6 targeting. Additionally, the interaction between RDR6 and the RDR6-interacting protein SGS3 involves liquid-liquid phase separation, suggesting its role in siRNA body formation.
