Summary: Inorganic polyphosphate is an ancient and ubiquitous polymer that is present in all organisms. While its role in bacteria and yeast has been described, its exact function in mammalian physiology remains poorly understood. Our study suggests that the metabolism of mitochondrial polyphosphate could be a potential pharmacological target in neurodegenerative disorders and neuronal aging.

Mitochondrial calcium uptake regulates tumour progression in embryonal rhabdomyosarcoma

Summary: Embryonal rhabdomyosarcoma (ERMS) is characterized by the elevated expression of mitochondrial calcium uniporter (MCU). Knockdown of MCU impairs mitochondrial calcium uptake and reduces mitochondrial reactive oxygen species (mROS) levels, leading to reduced cellular proliferation and motility, and increased differentiation. RNA sequencing reveals the involvement of MCU in TGF beta signaling, which plays significant roles in oncogenesis and myogenic differentiation. Modulation of mROS production impacts TGF beta signaling. Targeting the MCU complex to restore mitochondrial calcium homeostasis could be a potential therapeutic approach in ERMS.

Calcinosis in dermatomyositis: Origins and possible therapeutic avenues

Summary: This article discusses the pathogenic mechanisms and available treatments for calcinosis. There are currently no widely effective treatments, but various formulations of sodium thiosulfate show potential efficacy in calcinosis treatment. Early aggressive immunosuppression may prevent calcinosis in juvenile dermatomyositis, but the improvement of established calcinosis is limited. Surgical removal is a viable treatment option.
