Microalgae-Derived Pigments for the Food Industry

Summary: In the food industry, there is a growing focus on natural pigments instead of synthetic ones due to their excellent coloring ability and health properties. Microalgae are recognized as major sources of naturally derived commercial pigments and have gained significant value in the global food pigment market. This review provides updated information on the characteristics, application fields, production stability, and extraction processes of microalgae-derived pigments, particularly chlorophylls, carotenoids, and phycobiliproteins. The potential uses of these pigments as eco-friendly colorants, nutraceuticals, and antioxidants in food products are discussed, along with cultivation strategies and technical challenges in downstream processing.

An unexpected hydratase synthesizes the green light-absorbing pigment fucoxanthin

Summary: The ketocarotenoid fucoxanthin and its derivatives are able to absorb blue-green light prevalent in marine environments. In contrast to land plants, fucoxanthin is a primary light-harvesting pigment in phytoplankton species. However, the last steps of fucoxanthin biosynthesis have not been well understood until the discovery of the carotenoid isomerase-like protein CRTISO5. This protein functions as a fucoxanthin synthase in diatoms and has the unique ability to hydrate phaneroxanthin to form fucoxanthin.

Comparative Study Highlights the Potential of Spectral Deconvolution for Fucoxanthin Screening in Live Phaeodactylum tricornutum Cultures

Summary: Microalgal biotechnology has great potential in various industrial avenues but is limited by common processing methods that hinder high throughput screening and development of improved microalgal strains. In this study, we adapted a previously described method for monitoring fucoxanthin in Phaeodactylum tricornutum to live culture samples using a microplate reader, achieving a high correlation to HPLC. Adaptation to 384-well microplates and removal of the need to equalize cultures by density further increased the screening rate.

It was found that GSAAT2 plays a major role in ALA synthesis and that GluTR accumulation is higher in gsa2 mutants. A modeled structure of the protein complex indicated that GluTR-binding protein (GBP) mediates the stable association of GluTR and GSAAT, ensuring adequate ALA synthesis.

Transcriptome analysis revealed different metabolic and key regulatory genes were active at different growth phases. Genes such as ISPD and VDE were identified as critical for the regulation of fucoxanthin biosynthesis in P. tricornutum.

Rapid Sorting of Fucoxanthin-Producing Phaeodactylum tricornutum Mutants by Flow Cytometry

Summary: This study introduces a new method for screening mutants with high fucoxanthin content, successfully identifying a linear correlation between content and fluorescence intensity in the diatom model organism rich in fucoxanthin. By using flow cytometry, high-fucoxanthin-content mutants were sorted out, with a significant increase in fucoxanthin content compared to the wild type after 20 days of cultivation. This efficient, rapid, and high-throughput approach provides a valuable tool for screening fucoxanthin-overproducing mutants.
