Imaging of Reactive Astrogliosis by Positron Emission Tomography

Ryuichi Harada et al.

Summary: Neurodegenerative diseases are characterized by neuronal loss, gliosis, and the deposition of misfolded proteins. Imaging the glial response in vivo using PET combined with A beta and tau PET can provide insights into the disease process and aid in diagnosis and monitoring of therapeutic response.

APOEε4 potentiates the relationship between amyloid-β and tau pathologies

Joseph Therriault et al.

Summary: Through analysis of three independent samples of cognitively unimpaired, mild cognitive impairment, and Alzheimer's disease subjects, the study found that the interaction effect between APOE epsilon 4 and amyloid-beta was related to increased tau load in Alzheimer's disease-vulnerable regions. The interaction between one APOE epsilon 4 allele and amyloid-beta led to increased tau load, while the interaction between amyloid-beta and two APOE epsilon 4 alleles was associated with a more widespread pattern of tau aggregation.

APOE interacts with tau PET to influence memory independently of amyloid PET in older adults without dementia

Summary: The study revealed that APOE may interact with tau independently of Aβ, and elevated MTL tau levels have negative cognitive consequences in Aβ-negative ε4 carriers.

Association of APOE4 and Clinical Variability in Alzheimer Disease With the Pattern of Tau- and Amyloid-PET

Summary: Different clinical phenotypes in AD are associated with distinct patterns of tau but not amyloid pathology. Older age and APOE4 are risk factors for AD that also influence disease expression by promoting a more medial temporal lobe-predominant pattern of tau pathology.

Clinical validity of increased cortical binding of tau ligands of the THK family and PBB3 on PET as biomarkers for Alzheimer's disease in the context of a structured 5-phase development framework

Konstantinos Chiotis et al.

Summary: Experts evaluated the clinical validity of tau PET ligands of the THK family and PBB3, finding that THK PET ligands have good discriminative ability between healthy controls and AD patients, while there is no evidence supporting the diagnostic accuracy of PBB3. Although much work remains to be done to complete the aims of phases 2 and 3, the validation process for these tracers may not be completed due to off-target binding and the development of second-generation tracers.
