A comprehensive review of the therapeutic potential of citrus bioflavonoid hesperidin against lifestyle-related disorders

Aftab Ahmad et al.

Summary: The fruits of the citrus family are rich in flavonoids, which have superior antioxidant properties. Citrus bioactive components have the potential to be beneficial and have no adverse effects or cytotoxicity in healthy individuals. Hesperidin, a flavanone glycoside found in citrus fruit peel, has shown effective roles as an antioxidant and anti-inflammatory bioactive compound in biological systems. Different studies have investigated these roles of hesperidin.

Lipid pathway dysfunction is prevalent in patients with Parkinson's disease

Jasmin Galper et al.

Summary: There is a potential link between genetic risk factors for Parkinson's disease and lipid-related functions. The serum lipid profiles of Parkinson's disease patients and LRRK2 mutation carriers differ from those of controls, and alterations in sphingolipids and glycerolipids are also observed in the cerebrospinal fluid of these individuals. Furthermore, lipid dysregulation in Parkinson's disease mainly involves sphingolipid metabolism, insulin signaling, and mitochondrial function.

Neuroprotective Effects and Therapeutic Potential of the Citrus Flavonoid Hesperetin in Neurodegenerative Diseases

Jasmine A. Evans et al.

Summary: Neurodegenerative disorders are a serious health threat, and neuroinflammation and oxidative stress play critical roles in their onset, progression, and pathogenesis. Hesperetin, an aglycone of hesperidin found in citrus fruits, has been shown to have neuroprotective effects in experimental models of neurodegenerative diseases, and can modulate the release of cellular anti-inflammatory and antioxidative stress mediators.

Alpha-Synuclein-Specific Naturally Occurring Antibodies Inhibit Aggregation In Vitro and In Vivo

Anne K. Braczynski et al.

Summary: Parkinson's disease is associated with changes in the abundance of naturally occurring antibodies (nAbs) against alpha-synuclein (α Syn). In this study, nAbs-alpha Syn were found to specifically bind monomeric α Syn and inhibit its aggregation in vitro. Additionally, the addition of nAbs-alpha Syn to cultured cells affected intracellular α Syn aggregates.

Polyphenols-Based Nanosheets of Propolis Modulate Cytotoxic Amyloid Fibril Assembly of α-Synuclein

Yasin Rafiei et al.

Summary: Natural compound PFP nanosheets inhibit the amyloid aggregation of α-synuclein and redirect its aggregation towards nontoxic off-pathway oligomers. The binding of PFP nanosheets with α-synuclein amyloid fibrils demonstrates their potential anti-aggregation activity in neurodegenerative diseases.

Baicalein exhibits differential effects and mechanisms towards disruption of α-synuclein fibrils with different polymorphs

Yifei Yao et al.

Summary: This study reveals that the disruptive effects and modes of baicalein on alpha-synuclein fibrils are polymorphism-dependent. It can damage both WT and E46K/H50Q mutant alpha-synuclein fibrils, making it a potential drug candidate to alleviate the pathological process of Parkinson's disease.

Punnida Arjsri et al.

Summary: In this study, it was found that the Thai traditional medicinal plant Clerodendrum petasites S. Moore (C. petasites) and its active flavonoid hesperetin can attenuate inflammatory responses induced by the spike glycoprotein of SARS-CoV-2 by targeting the NLRP3 inflammasome and related pathways.

Hesperetin promotes longevity and delays aging via activation of Cisd2 in naturally aged mice

Chi-Hsiao Yeh et al.

Summary: This study found that hesperetin, a promising Cisd2 activator, when orally administered late in life, enhances Cisd2 expression and prolongs healthspan in old mice. Hesperetin functions mainly in a Cisd2-dependent manner to ameliorate age-related metabolic decline, body composition changes, glucose dysregulation, and organ senescence. Furthermore, a youthful transcriptome pattern is regained after hesperetin treatment during old age.

Neuroprotective effect of metformin on dopaminergic neurodegeneration and α-synuclein aggregation in C. elegans model of Parkinson's disease

Nada Saewanee et al.

Summary: The study demonstrated that metformin has significant neuroprotective effects on dopaminergic neurons and alpha-synuclein in a Parkinson's disease model, reducing neurodegeneration, inhibiting protein aggregation, and impacting gene expression, while not affecting the lifespan of the model organism. These findings suggest the potential therapeutic effects of metformin on Parkinson's disease, with further investigation needed to determine the precise mechanisms involved.

The food additive fast green FCF inhibits α-synuclein aggregation, disassembles mature fibrils and protects against amyloid-induced neurotoxicity

Fenghua Wang et al.

Summary: The food additive fast green FCF was found to inhibit alpha-synuclein fibrillogenesis and reduce cytotoxicity by disrupting the pentamer structure, indicating its potential as a drug candidate against amyloid fibril aggregation in Parkinson's disease. The interaction mechanism between fast green FCF and alpha-synuclein was elucidated through molecular dynamics simulations, revealing the disruption of pentamer structure and reduction of beta-sheet content. Electrostatic interactions, hydrogen bonds, and pi-pi interactions were identified as key factors in the binding of fast green FCF to alpha-synuclein.
