Detecting DNA methylation in the early stages of cancer is vital for disease diagnosis, but current methods are often time-consuming and costly. Therefore, the development of simple and sensitive methods for direct detection of DNA methylation without the need for molecular techniques is a hot topic in bioscience. This article provides an overview of optical biosensors (including fluorescence, FRET, SERs, and colorimetric) that have been used to detect DNA methylation. Various labeled and label-free reactions, as well as the impact of nanomaterials on detection sensitivity, are explored. Additionally, the advantages and disadvantages of each method are comprehensively discussed. The use of microfluidic devices for evaluating DNA methylation and smartphone-based DNA damage analysis is also examined.
